Effects of fullerenol nanoparticles and amifostine on radiation-induced tissue damages: Histopathological analysis

Radiation is an important modality in cancer treatment, and eighty percent of cancer patients need radiotherapy at some point during their clinical management. However, radiation-induced damage to normal tissues restricts the therapeutic doses of radiation that can be delivered to tumours and thereby limits the effectiveness of the treatment. The use of radioprotectors represents an obvious strategy to obtain better tumour control using a higher dose in radiotherapy. However, most of the synthetic radioprotective compounds studied have shown inadequate clinical efficacy owing to their inherent toxicity and high cost. Hence, the development of radioprotective agents with lower toxicity and an extended window of protection has attracted a great deal of attention, and the identification of alternative agents that are less toxic and highly effective is an absolute necessity. Recent studies have shown that alpha-2-macroglobulin (α 2 M) possesses radioprotective effects. α 2 M is a tetrameric, disulfide-rich plasma glycoprotein that functions as a non-selective inhibitor of different types of non-specific proteases and as a carrier of cytokines, growth factors, and hormones. α 2 M induces protein factors whose interplay underlies radioprotection, which supports the idea that α 2 M is the central effector of natural radioprotection in the rat. Pretreatment with α 2 M has also induced a significant reduction of irradiation-induced DNA damage and the complete restoration of liver and body weight. Mihailovic Mirjana et al. concluded that the radioprotection provided by α 2 M was in part mediated through cytoprotection of new blood cells produced in the bone marrow; these authors also indicated that an important aspect of the radioprotective effect of amifostine was the result of the induction of the endogenous cytoprotective capability of α 2 M. The radioprotective effects of α 2 M are possibly due to antioxidant, anti-fibrosis, and anti-inflammatory functions, as well as the maintenance of homeostasis, and enhancement of the DNA repair and cell recovery processes. This review is the first to summarise the observations and elucidate the possible mechanisms responsible for the beneficial effects of α 2 M. The lacunae in the existing knowledge and directions for future research are also addressed.

Alpha-2-macroglobulin as a radioprotective agent: a review.

To examine the protective potential of the Cotinus coggygria Scop. methanol extract, Wistar rats were treated with the hepatotoxic compound pyrogallol, which possesses a potent ability to generate free radicals and induce oxidative stress. The ability of the extract to counteract the oxidative stress was examined in rats that were injected with the extract intraperitoneally (500 mg·(kg body weight)–1) either 2 or 12 h before the pyrogallol treatment. The extract possesses a reducing activity in vitro and an ability to chelate the ferrous ion both in vivo and in vitro. Application of the extract prior to pyrogallol treatment led to a decrease in the levels of thiobarbituric acid-reactive substances, aspartate aminotransferase, and alanine aminotransferase, increased activities of antioxidant enzymes and attenuation of DNA damage, as well as increased Akt activity and inhibition of NF-κB protein expression. Extract of the plant Cotinus coggygria Scop. attenuates pyrogallol-induced hepatic oxidative stress in Wistar rats

Radiological incidents or terrorist attacks would likely expose civilians and military personnel to high doses of ionizing radiation, leading to the development of acute radiation syndrome. We examined the effectiveness of prophylactic administration of a developmental radiation countermeasure, γ-tocotrienol (GT3), in a total-body irradiation (TBI) mouse model. CD2F1 mice received GT3 24 h prior to 11 Gy cobalt-60 gamma-irradiation. This dose of radiation induces severe hematopoietic acute radiation syndrome and moderate gastrointestinal injury. GT3 provided 100% protection, while the vehicle control group had 100% mortality. Two-dimensional differential in-gel electrophoresis was followed by mass spectrometry and Ingenuity Pathway Analysis (IPA). Analysis revealed a change in expression of 18 proteins in response to TBI, and these changes were reversed with prophylactic treatment of GT3. IPA revealed a network of associated proteins involved in cellular movement, immune cell trafficking, and inflammatory response. Of particular interest, significant expression changes in beta-2-glycoprotein 1, alpha-1-acid glycoprotein 1, alpha-2-macroglobulin, complement C3, mannose-binding protein C, and major urinary protein 6 were noted after TBI and reversed with GT3 treatment. This study reports the untargeted approach, the network, and specific serum proteins which could be translated as biomarkers of both radiation injury and protection by countermeasures.

Gamma-tocotrienol, a radiation countermeasure, reverses proteomic changes in serum following total-body gamma irradiation in mice

The acute inflammatory response of the liver associated with chronic food restriction was examined in adult female and male Wistar rats. The changes in the levels of serum markers of liver injury, AST and ALT and the appearance of a serum marker of inflammation, the acute phase protein (APP) haptoglobin (Hp) were assessed following turpentine treatment of well-nourished (WN) controls and undernourished (UN) rats. Undernutrition was induced by food restriction during a six week period by offering chow equivalent to 50% of the normal food intake. In the female rats undernutrition significantly potentiated liver injury and increased their sensitivity to the toxic effects of turpentine, which was opposite to the results obtained for males. Differences in the basal levels of AST, ALT and Hp between females and males imply that the effects of chronic food restriction on protein synthesis in the liver are gender related.

The effect of chronic food restriction on liver acute phase protein response in female and male Wistar rats

The importance of alpha2-macroglobulin (alpha2M) in natural radioprotection was studied by examining its radioprotective effectiveness in rat models of exogenously and endogenously, preexposure-increased alpha2M. Radioprotective efficacy was ascertained by the postirradiation survival rate, the restoration of body weight, and the leukocyte count, which were monitored during a 4-week follow-up period. The results were compared with the effects of a pretreatment with the synthetic radioprotective agent amifostine (Ami), which provides 100% protection in rats whole-body-irradiated by x-rays given in a dose of 6.7 Gy (LD50/30). Raising the plasma concentration of alpha2M 15-fold in male rats by a single intraperitoneal injection of purified protein provided 100% survival of irradiated animals. Female rats on the 19th day of pregnancy with endogenously elevated levels of alpha2M displayed improved survival (80%) compared with untreated rats (50% survival). After alpha2M administration, the pregnant, irradiated rats exhibited 100% survival. In both males and pregnant females, alpha2M administration promoted body weight and leukocyte postirradiation recovery as in Ami-pretreated rats. These findings, together with our observation that Ami administration induced a 45-fold increase in alpha2M in the circulation, led us to conclude that alpha2M has an essential role in both natural and amifostine-mediated radioprotection in the rat.
