Streptomyces peucetius var. caesius, obtained from S. peucetius, the daunomycin producing microorganism, by mutagenic treatment, differs from the parent culture by the color of the vegetative and aerial mycelia and by its antibiotic, producing ability. S. peucetius var. caesius accumulates adriamycin in submerged and aerated culture on a medium containing glucose, brewer's yeast, and inorganic, salts both in shake flasks and in stirred fementers. Isolation of the product is performed by solvent extraction, chromatography on buffered cellulose columns, and crystallization as the hydrochloride. The new antitumor agent, adriamycin, is the 14-hydroxv derivative of daunomyein.

Fifty-one essential oils extracted from plants of known origin were tested for their antimicrobial activity against three bacteria, Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli and four yeasts, Torulopsis utilis, Schizosaccharomyces pombe, Candida albicans and Saccharomyces cerevisiae using the drop diffusion method. All showed antimicrobial activity against at least one of the micro-organisms. Following this preliminary screening, 13 essential oils showing antimicrobial activity against at least five of the micro-organisms were tested in the range 50 micrograms ml-1 to 500 micrograms ml-1 using broth micro dilution techniques with dimethylsulphoxide (DMSO) as a dispersing solvent. The concentration of most of the oils required for total inhibition of growth was > 500 micrograms ml-1. Further studies on the antimicrobial action of cinnamon oil in the range 10-150 micrograms ml-1 showed that 50-fold higher activity was found when no dispersing solvent was used.

Antimicrobial action of essential oils : the effect of dimethylsulphoxide on the activity of cinnamon oil

Plant phenolics are a structurally diverse group of compounds synthesized by all higher plants. The toxicity of phenolics and the products of their hydrolysis and oxidation has long been appreciated, and has implicated this group of natural products with plant defense. Unfortunately, much of the literature relating phenolics to plant defense is published in sources rarely consulted by evolutionists or ecologists. The purpose of this paper is to make a case for plant phenolics as defensive substances by considering their structural diversity, concentration, alteration upon infection, and mode of action. Correlations between ecogeographical and ecological parameters and phenolic profiles are discussed in light of the putative defensive role of phenolics, and additional correlations are predicted.

Plant Phenolics: An Ecological Perspective

Eugenol and trans-cinnamaldehyde are natural compounds known to be highly effective antimicrobials; however, both are hydrophobic molecules, a limitation to their use within the food industry. The goal of this study was to synthesize spherical poly (DL-lactide-co-glycolide) (PLGA) nanoparticles with entrapped eugenol and trans-cinnamaldehyde for future antimicrobial delivery applications. The emulsion evaporation method was used to form the nanoparticles in the presence of poly (vinyl alcohol) (PVA) as a surfactant. The inclusion of antimicrobial compounds into the PLGA nanoparticles was accomplished in the organic phase. Synthesis was followed by ultrafiltration (performed to eliminate the excess of PVA and antimicrobial compound) and freeze-drying. The nanoparticles were characterized by their shape, size, entrapment efficiency, and antimicrobial efficiency. The entrapment efficiency for eugenol and trans-cinnamaldehyde was approximately 98% and 92%, respectively. Controlled release experiments conducted in vitro at 37 °C and 100 rpm for 72 h showed an initial burst followed by a slower rate of release of the antimicrobial entrapped inside the PLGA matrix. All loaded nanoparticles formulations proved to be efficient in inhibiting growth of Salmonella spp. (Gram-negative bacterium) and Listeria spp. (Gram-positive bacterium) with concentrations ranging from 20 to 10 mg/mL. Results suggest that the application of these antimicrobial nanoparticles in food systems may be effective at inhibiting specific pathogens.

Practical Application:  Nanoencapsulation of lipophilic antimicrobial compounds has great potential for improving the effectiveness and efficiency of delivery in food systems. This study consisted of synthesizing PLGA nanoparticles with entrapped eugenol and trans-cinnamaldehyde. By characterizing these new delivery systems, one can understand the controlled-release mechanism and antimicrobial efficiency that provides a foundation that will enable food manufacturers to design smart food systems for future delivery applications, including packaging and processing, capable of ensuring food safety to consumers.

In 1799, Hatchett decalcified shells of crabs, lobsters, prawns and crayfish with mineral acids, observing that they produced a moderate effervescence and in a short time were found to be soft and plastic of a yellowish color and like a cartilage, which retained the original figure. Although this is the first mention of calcified chitin in invertebrates, the discovery of chitin is usually attributed both to Braconnot in 1811 who discovered chitin from fungi, and to Odier in 1823 who obtained a hornlike material after treatment of cockchafer elytra with potassium hydroxide. Chitin was first named fongine by Braconnot and then chitine by Odier. Children revealed the nitrogenous nature of chitin in 1824. The history of chitosan, the main derivative of chitin, dates back to 1859 with the work of Rouget. The name of chitosan was, however, introduced in 1894 by Hoppe-Seyler. In 1876, Ledderhose hydrolyzed arthropod chitin and discovered glykosamin, the first derivative of chitin. This review describes the 220 years of the development of chitin. I have roughly divided the story into five periods: discovery from 1799 to 1894, a period of confusion and controversy from 1894 to 1930, exploration in 1930–1950, a period of doubt from 1950 to 1970, and finally the period of application from 1970. The different periods are illustrated by examples of published studies, in particular from outstanding scholars who have left their mark on the history of this polysaccharide. Although this historic review is not exhaustive, it highlights the work of researchers who have contributed to the development of our knowledge of chitin throughout the 220 years of its history.
