Disruption of Pancreatic β-Cell Lipid Rafts Modifies Kv2.1 Channel Gating and Insulin Exocytosis
Fuzhen Xia,Xiaodong Gao,Edwin P. Kwan,Patrick P.L. Lam,Lillian Chan,Keiyan Sy,Laura Sheu,Michael B. Wheeler,Herbert Y. Gaisano,Robert G. Tsushima +9 more
TL;DR: Membrane compartmentalization of ion channels and SNARE proteins in lipid rafts may be critical for the temporal and spatial coordination of insulin release, forming what has been described as the excitosome complex.

Dual Role of VAMP8 in Regulating Insulin Exocytosis and Islet β Cell Growth
Dan Zhu,Yi Zhang,Patrick P.L. Lam,Subhankar Dolai,Yunfeng Liu,Erica P. Cai,Diana Choi,Stephanie A. Schroer,Youhou Kang,Emma M. Allister,Tairan Qin,Michael B. Wheeler,Cheng-Chun Wang,Wanjin Hong,Wanjin Hong,Minna Woo,Minna Woo,Herbert Y. Gaisano +17 more
TL;DR: Despite the β cell exocytotic defect, VAMP8-null mice have an increased total insulin secretory capacity, which improved glucose homeostasis, and it is concluded that these VAMP 8-mediated events partly underlie the therapeutic actions of GLP-1 on insulin secretion and β cell growth.

Transgenic Mouse Overexpressing Syntaxin-1A as a Diabetes Model
Patrick P.L. Lam,Yuk-Man Leung,Laura Sheu,James Ellis,Robert G. Tsushima,Lucy R. Osborne,Herbert Y. Gaisano +6 more
TL;DR: In this article, the specific role of syntaxin-1A in pancreatic beta-cells was investigated by generating transgenic mice, which expressed a moderately increased level (approximately 30% higher) of syntax-in-one-α (STX-1α) protein in the pancreatic islets.

Pancreatitis-Induced Depletion of Syntaxin 2 Promotes Autophagy and Increases Basolateral Exocytosis.
Subhankar Dolai,Tao Liang,Abrahim I. Orabi,Douglas Holmyard,Li Xie,Dafna Greitzer-Antes,Youhou Kang,Huanli Xie,Tanveer A. Javed,Patrick P.L. Lam,Deborah C. Rubin,Peter Thorn,Herbert Y. Gaisano +12 more
TL;DR: It is found that STX2 to block STX3- and STX4-mediated fusion of zymogen granules with the plasma membrane and exocytosis and prevent binding of ATG16L1 to clathrin, which contributes to induction of autophagy.

Munc18b Is a Major Mediator of Insulin Exocytosis in Rat Pancreatic β-Cells
Patrick P.L. Lam,Mitsuyo Ohno,Subhankar Dolai,Yu He,Tairan Qin,Tao Liang,Dan Zhu,Youhou Kang,Yunfeng Liu,Maria Kauppi,Li Xie,Wilson C.Y. Wan,Na-Rhum Bin,Na-Rhum Bin,Shuzo Sugita,Shuzo Sugita,Vesa M. Olkkonen,Noriko Takahashi,Haruo Kasai,Herbert Y. Gaisano +19 more
TL;DR: Munc18b-mediated exocytosis may be deployed to increase secretory efficiency of SGs in deeper cytosolic layers of β-cells as well as additional primary exocyTosis, which may open new avenues of therapy development for diabetes.
