Information coding in the olfactory system: Evidence for a stereotyped and highly organized epitope map in the olfactory bulb

Olfactory sensory neurons detect a large variety of odor molecules and send information through their axons to the olfactory bulb, the first site for the processing of olfactory information in the brain. The axonal connection is precisely organized so that signals from 1000 different types of odorant receptors are sorted out in 1800 glomeruli in the mouse olfactory bulb. Individual glomerular modules presumably represent a single type of receptor and are thus tuned to specific molecular features of odorants. Local neuronal circuits in the bulb mediate lateral inhibition among glomerular modules to sharpen the tuning specificity of output neurons. They also mediate synchronized oscillatory discharges among specific combinations of output neurons and may contribute to the integration of signals from distinct odorant receptors in the olfactory cortex.

The Olfactory Bulb: Coding and Processing of Odor Molecule Information

Candidate Drosophila olfactory receptors (ORs) provide molecular tools to investigate how the organization of the Drosophila olfactory system determines the coding of olfactory stimuli. Neurons in the third antennal segment and maxillary palp appear to express different ORs. Individual olfactory neurons send axonal projections to glomeruli in the antennal lobe. Using transgenic flies, we provide evidence that the neurons expressing a given OR gene, which have cell bodies distributed among neurons expressing other ORs, converge in their projections to topographically fixed glomeruli in the antennal lobe. This convergence allows for the formation of an odotopic map in the antennal lobe whose organization could provide a basis for olfactory discrimination in Drosophila.

A PCR-based strategy for amplifying putative receptors involved in murine olfaction was employed to isolate a member (OR3) of the seven-transmembrane-domain receptor superfamily During development, the first cells that express OR3 appear adjacent to the wall of the telencephalic vesicle at embryonic day 10 The OR3 receptor is uniquely expressed in a subset of olfactory cells that have a characteristic bilateral symmetry in the adult olfactory epithelium This receptor and its specific pattern of expression may serve a functional role in odor coding or, alternatively, may play a role in the development of the olfactory system
