Adherence of bacteria to hydrocarbons: A simple method for measuring cell‐surface hydrophobicity

ETEC is an underrecognized but extremely important cause of diarrhea in the developing world where there is inadequate clean water and poor sanitation. It is the most frequent bacterial cause of diarrhea in children and adults living in these areas and also the most common cause of traveler's diarrhea. ETEC diarrhea is most frequently seen in children, suggesting that a protective immune response occurs with age. The pathogenesis of ETEC-induced diarrhea is similar to that of cholera and includes the production of enterotoxins and colonization factors. The clinical symptoms of ETEC infection can range from mild diarrhea to a severe cholera-like syndrome. The effective treatment of ETEC diarrhea by rehydration is similar to treatment for cholera, but antibiotics are not used routinely for treatment except in traveler's diarrhea. The frequency and characterization of ETEC on a worldwide scale are inadequate because of the difficulty in recognizing the organisms; no simple diagnostic tests are presently available. Protection strategies, as for other enteric infections, include improvements in hygiene and development of effective vaccines. Increases in antimicrobial resistance will dictate the drugs used for the treatment of traveler's diarrhea. Efforts need to be made to improve our understanding of the worldwide importance of ETEC.

Enterotoxigenic escherichia coli in developing countries: epidemiology, microbiology, clinical features, treatment, and prevention

The ability of bacteria to attach to surfaces and develop into a biofilm has been of considerable interest to many groups in numerous industries, including the medical and food industry. However, little is understood in the critical initial step seen in all biofilm development, the initial bacterial cell attachment to a surface. This initial attachment is critical for the formation of a bacterial biofilm, as all other cells within a biofilm structure rely on the interaction between surface and bacterial cell for their survival. This review examines what are believed to be some of the most important aspects involved in bacterial attachment to a surface.

Host-specific fimbrial adhesins of noninvasive enterotoxigenic Escherichia coli strains.

Cell surface charge and hydrophobicity of Bacillus subtilis, Escherichia coli O157:H7, Listeria monocytogenes, Salmonella typhimurium, Serratia marcescens, Staphylococcus aureus, and Staphylococcus epidermidis were determined by hydrocarbon adherence, hydrophobic interaction, and electrostatic interaction chromatography. Surface charge and hydrophobicity were compared with the initial attachment values and rates of attachment of the bacteria to meat surfaces. There was a linear correlation between the relative negative charge on the bacterial cell surface and initial attachment to lean beef muscle (r2 = 0.885) and fat tissue (r2 = 0.777). Hydrophobicity correlated well with attachment to fat tissue only. The relative hydrophobicity of each bacterium was dependent on the specific method of determination, with wide variations noted between methods.

Porcine enteropathogenic Escherichia coli strains possessing or lacking K88 antigen were studied by using hydrophobic interaction chromatography on cross-linked agarose gels with alkyl or aryl substituents (amphiphilic gels) to determine whether or not they possessed surface-associated hydrophobic properties. Strains with K88ab or K88ac antigen adsorbed to phenyl and octyl Sepharose gels in the presence of 4 M sodium chloride. This property correlated with phenotypic expression of K88 antigen. Cells grown at 37 degrees C but not those grown at 18 degrees C possessed hydrophobic adsorptive characteristics in addition to the property of mannose-resistant hemagglutination of guinea pig erythrocytes. Adsorption of K88-positive strains to gels with hydrophobic ligands was independent of O group and enterotoxicity. Strains lacking K88 antigen did not adsorb to the hydrophobically substituted derivatives of Sepharose and lacked mannose-resistant hemagglutinating characteristics. Neither the presence of additional polysaccharide K antigens nor nonhemagglutinating pili conferred the property of hydrophobic interaction on the strains. K88-positive bacteria had a lower electrophoretic migration rate than did K88-negative bacteria of the same serotype in free-zone electrophoresis. K88-positive bacteria also adsorbed strongly to hydrophobic ligands in the presence of 1 M ammonium sulfate, whereas K88-negative strains did not. These observations provide evidence for the suspected role of hydrophobic interaction in the adhesive properties of certain enteropathogenic strains of E. coli. Moreover, hydrophobic interaction chromatography provides convenient and rapid alternative means of screening strains for a property potentially associated with adhesiveness.

All of 86 food routinely examined for potentially pathogenic enteric bacteria were found to harbour one or more coliform species. None of the strains isolated produced heat-labile enterotoxin (LT) or showed invasive properties. The suckling mouse test indicated that one strain of Escherichia coli produced heat-stable enterotoxin (ST). Twelve incidents of suspected food poisoning were also investigated. In two of them the foods examined contained LT-producing strains of E. coli and in two there were LT-producing strains of Klebsiella pneumoniae. The counts of viable enterotoxigenic micro-organisms in these foods were 3000-30,000 E. coli/g and 50,000 to 1 million K. pneumoniae/g. The dominant symptom in all the incidents was watery diarrhoea. These seem to be the first reported cases of foodborne enterotoxigenic enteric bacteria in Europe. Though enterotoxigenic E. coli and related gram-negative enterotoxin-producing species are rare in correctly handled food in Sweden, these micro-organisms should be searched for when outbreaks of food poisoning are investigated.
