Comparison of spontaneous and hormone-induced reproduction of burbot (Lota lota L.) under hatchery conditions

Comparative transcriptomic analysis brings new insights into the response to acute temperature acclimation in burbot (Lota lota lota)

Final oocyte maturation (FOM) model and artificial reproduction of burbot spawners (Lota lota) originating from the F1 generation of a cultured stock in comparison to wild stock

Testis development and ultrastructure of spermatogenic cells and spermatozoa of burbot Lota lota, a commercially important cold freshwater fish, were studied by light and transmission electron microscopy. Spermatogonia, spermatocytes, spermatids, and spermatozoa are distributed along the seminiferous tubules. Electron-dense bodies appear in germ cells from primary spermatogonia to secondary spermatocytes. We identified three distinct stages of spermatid cell differentiation based on chromatin condensation, development of the flagellum, formation of a nuclear fossa, and elimination of excess cytoplasm. Spermatozoa were anacrosomal and characterized by location of the centrioles outside the nuclear fossa and incomplete perpendicular arrangement of the centrioles. The sperm flagellum displayed an axoneme with nine doublets of peripheral microtubules and two central microtubules. These results provide valuable information for burbot taxonomy and may clarify the process of spermatogenesis for this species.

Zusatzlich waren Trockengewicht, Cortisolgehalt und Grose der Eier mit der Fekunditat und dem Entwicklungspotential der Eier verknupft. Ein erheblicher Anteil der Variabilitat in der Eientwicklung (47.1% Befruchtung; 58.2% 24 h, 47% 48 h, 43.9% 72 h Embryouberleben; 46.6% Schlupf; 88.9% geschlupfte Larven) konnte durch eine Kombination von maternalen Merkmalen und Eiparametern erklart werden. Obwohl ein hoher Befruchtungserfolg (durchschnittlich 89.2%) verzeichnet wurde, gibt es Moglichkeiten um Befruchtungsprotokolle hinsichtlich der Handhabung mannlicher Gameten beim Zander zu optimieren. Die Spermien verschiedener Mannchen werden haufig vor der Befruchtung zusammengefuhrt (Pooling) oder kurz Zeit (Stunden) gelagert bis ovulierte Eier zur Verfugung stehen. Ein Verlust der Spermienqualitat (Motilitat, Geschwindigkeit) wahrend kurzer Lagerung konnte durch Puffer oder die Zugabe von Melatonin bzw. Progesteron nicht verhindert werden. Die Transfusion von Seminalflussigkeit zwischen Spermien mit niedriger und hoher Qualitat resultierte in einer Abnahme bei Mannchen mit anfanglich hoher Spermiengeschwindigkeit. Die Qualitat von Spermien mit niedriger Qualitat wurde nicht gesteigert. Diese Erkenntnisse liefern Empfehlungen fur das Management von Brutbetrieben und zeigen Potential zur Optimierung von Reproduktionsprotokollen bezuglich der in vitro Befruchtung beim Zander auf.%%%%The pikeperch (Sander lucioperca) has great potential for European aquaculture diversification, but the propagation is impeded by a shortage of stocking material caused by variability in gamete quality. Here, maternal size rather than out-of-season spawning or spawning experience was associated with variability in egg quality with very large females showing decreasing reproductive performance. Maternal factors were linked to the egg composition mainly in regard to fatty acid (FA) profiles, which in turn were correlated with rates of fertilization and embryo survival. In addition, egg size, dry weight and cortisol content were associated with fecundity and embryo survival until hatching. Reproductive management and gamete quality in pikeperch (Sander lucioperca)

A rearing experiment was conducted to test whether temperature protocols that differed from a simulation of natural conditions might induce maturation after isothermal grow-out in burbot Lota lota. Lota lota were acclimated to two different temperature regimes: low temperature (LT), close to natural temperature at 4·0° C and elevated, high temperature (HT) at 8·5° C over 40 and 27 days respectively, with all fish then wintered for 47 days. Every second fish was treated with a gonadotropin-releasing hormone analogue. Maturational competence of oocytes was assessed with a germinal vesicle breakdown assay using a novel staining strategy. In both treatments, puberty and maturational progress occurred, characterised by an elevated gonado-somatic index and advanced gonadal stages (histological analysis). Progress of gonadal maturation was reflected by elevated plasma concentrations of testosterone and 11-ketosterone in males and 17β-oestradiol in females. Ovulation was not observed. Sperm could be activated equally across treatments. In general, LT was more effective than HT treatment, indicated by advanced gonadal stages, higher numbers of oocytes undergoing germinal vesicle breakdown in vitro and elevated sex steroid levels. Hormone treatment could improve effectiveness at HT. In conclusion, less drastic temperature regimes as previously reported in combination with hormone treatments seem sufficient to induce maturation in L. lota after isothermal grow-out.
