The transcription factor Oct‐4 is expressed specifically in the totipotent germline cycle of mice. Cells that lose Oct‐4 differentiate along different paths to form embryonic and extraembryonic somatic tissue. Oct‐4 may maintain the potency of stem and germline cells by preventing all other differentiation pathways. Oct‐4 may also regulate the molecular differentiation of cells in the germ lineage as it progresses from the fertilized egg, through cleavage stage/morula blastomeres, blastocyst, inner cell mass, epiblast, germ cells, and gametes. The factors that regulate, and are regulated by, Oct‐4 are reviewed with respect to the phenomena of cell potency and germ/soma segregation and differentiation. Faced with the avalanche of genomic sequences and data on messenger RNA expression, biological scientists are confronting a frightening prospect: piles of information but only flakes of knowledge. How can the thousands of sequences being determined and deposited, and the thousands of expression profiles being generated by the new array methods, be synthesized into useful knowledge? What form will this knowledge take? These are questions being addressed by scientists in the field known as ‘functional genomics’.

Protein Function in the Post-Genomic Era

a small animal model such as rabbit to serve as a biological model for in vivo tissue engineering experiments, would be desirable. The aim of our preliminary study was to describe a method of isolation of rabbit multipotent stem cells from the amniotic fluid and to examine the phenotype of these cells using flow cytometry. new Zealand White rabbit amniotic fluid was mixed with the culture medium and transferred into a T25 tissue culture flask. analysis of rabbit stem cells was realized using specific antibodies and flow cytometry (BD FaCSCanto II flow cytometry analyzer). Our preliminary resutls show rapid proliferation of amniotic fluid-derived adherent cells. We hypothesize that these cells have a broadly multipotent mesenchymal stem cell phenotype basing on their morphology and the expression of CD44. Furthermore, these cells can potentially serve as an excellent and amenable cell source for reprogramming experiments into naive induced pluripotent stem cells.

Rabbit amniotic fluid as a potential alternative source of broadly multipotent stem cells

Embryonic stem cells bona fide encompass cells of the inner cell mass, explanted from blastocysts, that exist exclusively in in vitro culture systems. To qualify for a stem cell, one has to be capable of self-renewal, multilineage differentiation and functional repopulation of a target tissue. Besides essential aspects of stem cell biology, in this review, we discuss categories according to the differentiation potential or plasticity of stem cells which is closely intertwined with their developmental rank in mammals ranging from totipotent zygote to monopotent stem cells. Aspects of embryonic stem cells, their isolation, signalling pathways governing their self-renewal and pluripotency, markers, genetic modification and differentiation are reviewed. Finally, we outline some hurdles, challenges and outlooks in the field of stem cell research and its promises for regenerative and replacement therapies.
