Glycolipids, consisting of a carbohydrate moiety linked to fatty acids, are microbial surface active compounds produced by various microorganisms. They are characterized by high structural diversity and have the ability to decrease the surface and interfacial tension at the surface and interface, respectively. Rhamnolipids, trehalolipids, mannosylerythritol lipids and cellobiose lipids are among the most popular glycolipids. They have received much practical attention as biopesticides for controlling plant diseases and protecting stored products. As a result of their antifungal activity towards phytopathogenic fungi and larvicidal and mosquitocidal potencies, glycolipid biosurfactants permit the preservation of plants and plant crops from pest invasion. Also, as a result of their emulsifying and antibacterial activities, glycolipids have great potential as food additives and food preservatives. Furthermore, the valorization of food byproducts via the production of glycolipid biosurfactant has received much attention because it permits the bioconversion of byproducts on valuable compounds and decreases the cost of production. Generally, the use of glycolipids in many fields requires their retention from fermentation media. Accordingly, different strategies have been developed to extract and purify glycolipids. © 2016 Society of Chemical Industry.

Glycolipid biosurfactants: main properties and potential applications in agriculture and food industry

The role of lipopeptide biosurfactant on microbial remediation of aged polycyclic aromatic hydrocarbons (PAHs)-contaminated soil

Increasing concerns over limited petroleum resources and associated environmental problems are motivating the development of efficient cell factories to produce chemicals, fuels, and materials from renewable resources in an environmentally sustainable economical manner. Bacillus spp., the best characterized Gram-positive bacteria, possesses unique advantages as a host for producing microbial enzymes and industrially important biochemicals. With appropriate modifications to heterologous protein expression and metabolic engineering, Bacillus species are favorable industrial candidates for efficiently converting renewable resources to microbial enzymes, fine chemicals, bulk chemicals, and fuels. Here, we summarize the recent advances in developing Bacillus spp. as a cell factory. We review the available genetic tools, engineering strategies, genome sequence, genome-scale structure models, proteome, and secretion pathways, and we list successful examples of enzymes and industrially important biochemicals produced by Bacillus spp. Furthermore, we highlight the limitations and challenges in developing Bacillus spp. as a robust and efficient production host, and we discuss in the context of systems and synthetic biology the emerging opportunities and future research prospects in developing Bacillus spp. as a microbial cell factory.

Glycolipids are characterized by highly structural diversity and have the ability to decrease the surface and interfacial tension at the surface and interface respectively. It presented initially a detailed classification of glycolipid including rhamnolipids, trehalolipids, mannosylerythritol-lipids, cellobiolipids; along with their producing strain. The review described the main functional properties of glycolipid including emulsification/de-emulsification capacity, foaming and moisturizing, viscosity reduction and hydrocarbon solubilizing and mobilizing capacities. Owing these properties, they can be applied in environmental fields as hydrocarbon emulsifiers, solubilizing and mobilizing agents, for their moisturizing capabilities and ability to reduce viscosity. The review will present a detailed classification of glycolipid biosurfactants, functional properties and the potential related applications in environment and bioremediation.

Glycolipid Biosurfactants, Main Classes, Functional Properties and Related Potential Applications in Environmental Biotechnology

Globally the change towards the establishment of a bio-based economy has resulted in an increased need for bio-based applications. This, in turn, has served as a driving force for the discovery and application of novel biosurfactants. The class Actinobacteria represents a vast group of microorganisms with the ability to produce a diverse range of secondary metabolites, including surfactants. Understanding the extensive nature of the biosurfactants produced by actinobacterial strains can assist in finding novel biosurfactants with new potential applications. This review therefore presents a comprehensive overview of the knowledge available on actinobacterial surfactants, the chemical structures that have been completely or partly elucidated, as well as the identity of the biosurfactant-producing strains. Producer strains of not yet elucidated compounds are discussed, as well as the original habitats of all the producer strains, which seems to indicate that biosurfactant production is environmentally driven. Methodology applied in the isolation, purification and structural elucidation of the different types of surface active compounds, as well as surfactant activity tests, are also discussed. Overall, actinobacterial surfactants can be summarized to include the dominantly occurring trehalose-comprising surfactants, other non-trehalose containing glycolipids, lipopeptides and the more rare actinobacterial surfactants. The lack of structural information on a large proportion of actinobacterial surfactants should be considered as a driving force to further explore the abundance and diversity of these compounds. This would allow for a better understanding of actinobacterial surface active compounds and their potential for biotechnological application.

Surfactants tailored by the class Actinobacteria

Biosurfactant-producing bacteria were isolated from mangrove sediment in southern Thailand. Isolates were screened for biosurfactant production by using the surface tension test. The highest reduction of surface tension was achieved with a bacterial strain which was identified by 16S rRNA gene sequencing as Oleomonas sagaranensis AT18. It has also been investigated using different carbon and nitrogen sources. It showed that the strain was able to grow and reduce the surface tension of the culture supernatant to 25 mN/m. In all 5.30 g of biosurfactant yield was obtained after 54 h of cultivation by using molasses and NaNO3 as carbon and nitrogen sources, respectively. The biosurfactant recovery by chloroform:methanol extraction showed a small critical micelle concentration value (8 mg/l), thermal and pH stability with respect to surface tension reduction. It also showed emulsification activity and a high level of salt concentration. The biosurfactant obtained was confirmed as a glycolipid by using a biochemical test, FT-IR and mass spectra. The crude biosurfactant showed a broad spectrum of antimicrobial activity and also had the ability to emulsify oil and enhance PAHs solubility.

Isolation and functional characterization of a biosurfactant produced by a new and promising strain of Oleomonas sagaranensis AT18

Microbial surface active compounds (SACs) were isolated from various environmental sources in Thailand. Isolates were screened for SACs production in different carbon sources (crude glycerol, commercial sugar, decanter, glucose, molasses, used palm oil, and used lubricating oil) by using drop-collapsing test and emulsification activity. Molasses produced the highest number of positive results (23 of 25 isolates). Twenty-one isolate strains produced emulsions with xylene, and 15 exhibited high emulsion-stabilizing capacity, maintaining more than 50 % of the original emulsion volume for 24 h, and six isolate strains reduced the growth medium surface tension to 40 mN/m. The phylogenetic position of these 25 isolates was evaluated by 16S rRNA gene sequence analysis. The production of microbial SACs was determined for strains representative of 16 different bacterial genera, in which ten genera (Blastococcus, Erysipelothrix, Humicoccus, Methylophilus, Microlunatus, Nevskia, Pectinatus, Rubrimonas, Selenomonas, and Stenotrophomonas) were firstly reported as SAC-producing strains. Overall, the new SAC-producing strains isolated in this study display promising features for the future development and use in economically efficient industrial-scale biotechnological processes.

Characterization and phylogenetic analysis of microbial surface active compound-producing bacteria.

This study presents isolation of biosurfactant (BS)-producing bacteria, Bacillus subtilis TD4 (BsTD4) and Pseudomonas aeruginosa SU7 (PaSU7), from palm oil contaminated soil. BS production was done by using a minimal salt medium with used crude glycerol as carbon and commercial monosodium glutamate as nitrogen source. After microbial cultivations at 30°C under optimum conditions, BSs produced by BsTD4 and PaSU7 reduced surface tension of culture supernatant to 30 mN/m with BS yield of 2.65 and 2.25 g/l, respectively. BSs could reduce surface tension of pure water to 26.0 and 28.0 mN/m with critical micelle concentration (CMC) of 12 mg/l (BsTD4) and 20 mg/l (PaSU7). BS produced by BsTD4 exhibited higher oil recovery efficiency than that produced by PaSU7. Both BSs could recover oil more effectively than synthetic surfactants, could enhance solubility of PAHs, and can emulsify various oil and hydrophobic compounds.
