Recent advances of iridium(III) metallophosphors for health-related applications

In this paper, a new kind of colorimetric receptors R1 and R2 for fluoride ions have been synthesized and characterized by various spectral techniques. Single crystal X-ray diffraction study confirms, the receptor R2 possesses the orthorhombic centrosymmetric lattice with the donor atoms of ONS. In naked eye, absorption and fluorescence studies were evidently predicting the potential sensing capability of the receptors R1 and R2 that detect fluoride ions by competing with various ions. The detection limit was found to be more or less same as low as 10 −7  M for R1 and R2 with F − . We proposed the 1:1 binding formation of R1 and R2 by means of F − ion was further supported by NMR titration and Job’s plot analysis. Besides, the density functional theory (DFT) calculations have been theoretically sustained with the sensing mechanism. Moreover, both the two receptors had the possibility of potentially applied in an analytical application for detecting the trace amounts of fluoride ion in the real water sample analysis was demonstrated. Live cell imaging experiments with the receptors displayed that these receptors extensively applicable to detect fluoride ions in HeLa cells under physiological conditions. Finally, the use of test strips containing R1 and R2 that sense fluoride ions in solution medium and supported on solid medium (silica) was exhibited.

Phosphorescent Ir(III) complexes are expected to be new multifunctional theranostic platforms that enable the integration of imaging capabilities and anticancer properties. Mitophagy is an important selective autophagic process that degrades dysfunctional mitochondria. Until now, the regulation of mitophagy is still poorly understood. Herein, we present two phosphorescent cyclometalated iridium(III) complexes (Ir1 and Ir2) that can accumulate in mitochondria and induce mitophagy. Because of their intrinsic phosphorescence, they can specially image mitochondria and track mitochondrial morphological alterations. Mechanism studies show that Ir1 and Ir2 induce mitophagy by depolarization of mitochondrial membrane potential, depletion of cellular ATP, perturbation in mitochondrial metabolic status, and induction of oxidative stress. Shining New Light on Biological Systems: Luminescent Transition Metal Complexes for Bioimaging and Biosensing Applications

Two new biscyclometalated complexes [Ir(ptzR)2(dppz)]+ (dppz = dipyridophenazene; ptzRH = 4-phenyl-1-benzyl-1,2,3-triazole (1+) and 4-phenyl-1-propyl-1,2,3-triazole (2+)) have been prepared. The hexafluorophosphate salts of these complexes have been fully characterized and, in one case, the X-ray structure of a nitrate salt was obtained. The DNA binding properties of the chloride salts of the complexes were investigated, as well as their cellular uptake by A2780 and MCF7 cell lines. Both complexes display an increase in the intensity of phosphorescence upon titration with duplex DNA, indicating the intercalation of the dppz ligand and, given that they are monocations, the complexes exhibit appreciable DNA binding affinity. Optical microscopy studies reveal that both complexes are taken up by live cancer cell lines displaying cytosol based luminescence. Colocalization studies with commercial probes show high Pearson coefficients with mitotracker dyes confirming that the new complexes specifically localize on mitochondria.

Mitochondria-localising DNA-binding biscyclometalated phenyltriazole iridium(III) dipyridophenazene complexes: syntheses and cellular imaging properties

A reducing intracellular environment is necessary for living cells. Here a redox-sensitive phosphorescent probe Ir-NO has been developed for evaluating the redox environment in living cells. Upon addition of reducing molecules, such as glutathione and ascorbic acid, the phosphorescent intensity of the probe is turned on, and the emission lifetime is elongated evidently. Furthermore, this probe has been used for optical imaging of the intracellular reducing environment by utilizing confocal laser scanning microscopy and phosphorescence lifetime imaging microscopy.
