Filamins are large actin-binding proteins that stabilize delicate three-dimensional actin webs and link them to cellular membranes. They integrate cellular architectural and signalling functions and are essential for fetal development and cell locomotion. Here, we describe the history, structure and function of this group of proteins.

Filamins are a family of high molecular mass cytoskeletal proteins that organize filamentous actin in networks and stress fibers. Over the past few years it has become clear that filamins anchor various transmembrane proteins to the actin cytoskeleton and provide a scaffold for a wide range of cytoplasmic signaling proteins. The recent cloning of three human filamins and studies on filamin orthologues from chicken and Drosophila revealed unexpected complexity of the filamin family, the biological implications of which have just started to be addressed. Expression of dysfunctional filamin-A leads to the genetic disorder of ventricular heterotopia and gives reason to expect that abnormalities in the other isogenes may also be connected with human disease. Structural and functional aspects of ¢lamins

Objective. Systemic lupus erythematosus (SLE) is associated with high cardiovascular morbidity and mortality. Cardiovascular involvement is frequently underestimated by routine imaging techniques. Our aim was to determine if new echocardiographic imaging modalities like tissue Doppler (TDI), strain rate (SRR), and strain (SRI) imaging detect abnormalities in left ventricular (LV) function in asymptomatic patients with SLE. Methods. Sixty-seven young patients with SLE (mean age 42 ± 10 yrs) without typical symptoms or signs of heart failure or angina, and a matched healthy control group (n = 40), underwent standard transthoracic echocardiography, TDI, SRR, and SRI imaging of the LV as well as assessment of disease characteristics. Results. Despite findings within the normal range on routine standard 2-dimensional echocardiography, SLE was associated with significantly impaired systolic and diastolic myocardial velocities of the LV measured by TDI [mean global TDI: systolic (s): 2.9 ± 0.9 vs 3.9 ± 0.7 cm/s, p Conclusion. SLE is associated with a significant impairment of systolic and diastolic LV longitudinal function in patients without cardiac symptoms. New imaging modalities provide earlier insight into cardiovascular involvement in SLE and seem to be superior to standard echocardiography to detect subclinical myocardial disease.

Myocardial left ventricular dysfunction in patients with systemic lupus erythematosus: new insights from tissue Doppler and strain imaging

The platelet cytoskeleton, a system of actin and tubulin polymers, maintains the discoid structure of the resting platelet as it circulates in blood. Tubulin is assembled into microtubules that are organized into a marginal ring and confer the discoid shape on the platelet. To change shape following activation, platelets disassemble their spectrin-based membrane cytoskeleton and its underlying actin filaments and then reassemble new cortical actin filaments to spread over surfaces and to generate filopods. Actin assembly is regulated by numerous actin binding proteins. Microtubules are reorganized during platelet actin and shape change. The microtubule coil is compressed into the cell center, disassembled, and individual microtubule are sent from the cell center into the various platelet projections.

Human filamin gene FLNB encodes a protein which interacts with the cytoplasmic tail of glycoprotein Ibalpha. Here we report on the mapping of the FLNB gene coding region to chromosome region 3p21.1→p14.3 by fluorescence in situ hybridization with YACs from a previously established 3p14–specific YAC contig. By establishing STSs for 5)and 3)-end-sequences of the FLNB coding region, FLNB was localized centromeric to the translocation breakpoint t(3;6) in 3p14.3 and distal to the translocation breakpoint t(3;8) in 3p14.2 by STS-PCR. Together these results show that the FLNB coding region maps to chromosome region 3p14.3. Furthermore, by STS-PCR analysis two YACs were identified which contain the entire transcribed region of the FLNB gene.
