Cellular dynamics following CAR T cell therapy are associated with response and toxicity in relapsed/refractory myeloma.
Luise Fischer,N. Grieb,Patrick Born,Ronald Weiss,Sabine Seiffert,Andreas Boldt,Stephan Fricke,Paul Franz,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,Heike Weidner,S. Wang,Enrica Bach,Sandra Hoffmann,J. Ussmann,Janine Kirchberg,Saskia Hell,Sebastian Schwind,K. Metzeler,M. Herling,Madlen Jentzsch,Georg-Nikolaus Franke,Ulrich Sack,Kristin Reiche,Ulrike Köhl,Uwe Platzbecker,Vladan Vucinic,Maximilian Merz +28 more
TL;DR: The results show that in vivo expansion of CD8+ CAR T cells is correlated to response, however persistence is not essential for durable remission in RRMM patients, and provide evidence, that an increased fraction of CD8+ T cells at day of leukapheresis in combination with successful lymphodepletion positively influence the outcome.

A digital twin model for evidence-based clinical decision support in multiple myeloma treatment
N. Grieb,L. Schmierer,Sarah Strobel,Christian Schulz,T. Meschke,Anne Sophie Kubasch,Annamaria Brioli,Uwe Platzbecker,Thomas Neumuth,Maximilian Merz,Alexander Oeser +10 more
TL;DR: This work proposes a collaborative approach that mitigates the regulatory and ethical concerns that are broadly discussed when automated decision-making tools are to be included into clinical routine and defines an initial evaluation scenario in the context of patient characterization and treatment outcome simulation as an exemplary use case for the MMDT.

Activation of distinct inflammatory pathways in subgroups of LR-MDS.
Marie Schneider,C.W. Rolfs,Matt Trumpp,Susann Winter,Luise Fischer,Mandy Richter,Victoria Menger,Kolja Nenoff,N. Grieb,Klaus H. Metzeler,Anne Sophie Kubasch,Katja Sockel,Christian Thiede,Jincheng Wu,Jang Hee Woo,A. Brüderle,Lorenz C. Hofbauer,Andreas Roth,Michael Cross,Uwe Platzbecker +19 more
TL;DR: In this article , a large, clinically and genetically well-characterized cohort of treatment-naïve MDS patients confirms intrinsic activation of inflammatory pathways in general mediated by caspase-1, interleukin (IL)-1β and IL-18 in low risk (LR)-MDS bone marrow and reveals a previously unrecognized heterogeneity of inflammation between genetically defined LR-MDS subgroups.
Single-cell multiomic dissection of response and resistance to chimeric antigen receptor T cells against BCMA in relapsed multiple myeloma.
Michael Rade,N. Grieb,Ronald Weiss,Jaren Sia,Luise Fischer,Patrick Born,Andreas Boldt,Stephan Fricke,Paul Franz,Jonathan Scolnick,Lakshmi Venkatraman,Stacy Xu,Christina Kloetzer,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,S. Wang,Enrica Bach,Sandra Hoffmann,J. Ussmann,B. Schetschorke,Saskia Hell,Sebastian Schwind,K. Metzeler,Marco Herling,Madlen Jentzsch,G.-N. Franke,Ulrich Sack,Ulrike Köhl,U. Platzbecker,Kristin Reiche,Vladan Vucinic,M. Merz +32 more
TL;DR: This work subjected mononuclear cells isolated from peripheral blood and bone marrow before and after the application of approved B cell maturation antigen-directed CAR T cells to single-cell multiomic analyses to identify markers associated with resistance and early relapse.
