White spot syndrome virus (WSSV) is a devastating pathogen in shrimp aquaculture. Standardized challenge procedures using a known amount of infectious virus would assist in evaluating strategies to reduce its impact. In this study, the shrimp infectious dose 50% endpoint (SID50 ml(-1)) of a Thai isolate of WSSV was determined by intramuscular inoculation (i.m.) in 60 and 135 d old specific pathogen-free (SPF) Litopenaeus vannamei using indirect immunofluorescence (IIF) and 1-step polymerase chain reaction (PCR). Also, the lethal dose 50% endpoint (LD50 ml(-1)) was determined from the proportion of dead shrimp. The median virus infection titers in 60 and 135 d old juveniles were 10(6.8) and 10(6.5) SID50 ml(-1), respectively. These titers were not significantly different (p > or = 0.05). The titration of the WSSV stock by oral intubation in 80 d old juveniles resulted in approximately 10-fold reduction in virus titer compared to i.m. inoculation. This lower titer is probably the result of physical and chemical barriers in the digestive tract of shrimp that hinder WSSV infectivity. The titers determined by infection were identical to the titers determined by mortality in all experiments using both i.m. and oral routes at 120 h post inoculation (hpi), indicating that every infected shrimp died. The determination of WSSV titers for dilutions administered by i.m. and oral routes constitutes the first step towards the standardization of challenge procedures to evaluate strategies to reduce WSSV infection.

In vivo titration of white spot syndrome virus (WSSV)

We screened the gill and somatic muscle of 152 wild-caught invasive giant tiger prawns (GTPs), Penaeus monodon Fabricius, 1798 (Penaeoidea: Penaeidae) for infection by white spot syndrome virus (WSSV), infectious hypodermal and hematopoietic necrosis virus (IHHNV), and Taura syndrome virus (TSV) using molecular methods (PCR and qPCR) and transmission electron microscopy (TEM). The sampled GTPs comprised 1 freshly-collected specimen from Mississippi Sound (Northern Gulf of Mexico) during 2020; 54 frozen specimens originally captured from the Northern Gulf of Mexico off Mississippi, Alabama, and Florida during 2014–2016; 76 frozen specimens originally captured from the Northwestern Atlantic Ocean off North Carolina, South Carolina, Georgia, and Florida during 2014–2020; and 21 museum-accessioned specimens (19 initially ethanol-preserved; 2 initially formalin-fixed) captured from the Gulf of Mexico and Northwestern Atlantic Ocean off Florida during 1988, 2011–2013, and 2016. Molecular viral detection relied upon qPCR with TaqMan chemistry for WSSV, conventional PCR for IHHNV, and rt-PCR for Taura virus. TEM was performed on WSSV qPCR+ positive GTP gill to confirm viral infection. A total of 18 GTPs were positive for WSSV by qPCR, 1 was positive for IHHNV by conventional PCR, and none were positive for Taura virus. This is the first report of a WSSV or IHHNV infection in a wild-caught GTP from the Gulf of Mexico or Northwestern Atlantic Ocean and first detection of an IHHNV infection in a wild-caught host in the Northwestern Atlantic Ocean. The phylogenetic analyses indicated that, broadly, sympatric WSSV isolates (unless identical) do not share a recent common ancestor (they are paraphyletic), suggesting that the virus has been repeatedly translocated and introduced into the Gulf of Mexico and Northwestern Atlantic Ocean and that it originated from different localities.

White spot syndrome virus (WSSV) is a virulent disease that disrupts shrimp farm operations throughout the world. While the United States has had only limited outbreaks of WSSV within the past several decades, it is important to ensure that this disease does not infect wild penaeid shrimp populations. In Texas, there is a potential for WSSV to spread to wild penaeid populations in the Gulf of Mexico via infected imported nonnative bait shrimp, imported broodstock, or wild crustacean hosts. Due to these potential threats, the Texas Parks and Wildlife Coastal Fisheries Division monitored WSSV in wild brown shrimp Farfantepenaeus aztecus and white shrimp Litopenaeus setiferus from seven major bay systems along the Texas coast during 2019. While no positive samples were detected from the collected shrimp, a power analysis illustrated a potential for low-level WSSV prevalence within Texas shrimp populations that would not be detectable by this monitoring survey. Overall, WSSV does not appear to be a major threat in the Texas region of the Gulf of Mexico, but continual observation and monitoring of wild penaeid shrimp is necessary to protect this resource from future WSSV outbreaks.

Diagnostic Molecular Investigation of White Spot Syndrome Virus Finds No Infection in Wild White Shrimp and Brown Shrimp along the Texas Gulf Coast.

Lastly, since VfmE binds c-di-GMP via the RxxxR motif, mutation in R93D of VfmE eliminated the c-di-GMP-related phenotypes in Pel activity. Our results demonstrate that VfmE not only regulates the Vfm quorum-sensing but also is a c-di-GMP effector, suggesting that D. dadantii integrates the c-di-GMP signaling network with the Vfm quorum-sensing pathway during environmental adaptation thereby regulating its virulence. between wild-type and the myh9b mutant zebrafish at the time points investigated. Upon ligand binding, EGL-15 activates signal transduction by the RAS-RAF-MEK-ERK pathway that has been implicated in many human diseases. EGL-15 is negatively regulated by CLR-1, a receptor tyrosine phosphatase. In the hypodermis, loss of clr-1 activity causes a fluid balance defect termed the “clear” (Clr) phenotype. EGL -15 dysregulation also results in defects that include the hyperactive Clr phenotype, as well as a “suppressor of clear” (Soc) phenotype, caused by mutations that compromise EGL-15 activity. We also describe a novel adaptor protein denoted SOC-3 that may offer an indirect coupling pathway towards ERK activation. The opportunistic pathogen, Pseudomonas aeruginosa, relies on the expression of type IV pili (T4P) for aspects of its virulence. T4P are long, flexible appendages used by bacteria in surface adhesion, biofilm maturation, and twitching motility. Expression of T4P-related genes and modulation of T4P activity in P. aeruginosa is dependent on the Chp chemosensory system. The core signaling complex of the Chp system consists of a single methyl-accepting chemotaxis protein (MCP; PilJ), two adaptor proteins (PilI, ChpC), and a histidine kinase (ChpA). These proteins organize themselves into large, highly ordered chemosensory arrays that are localized towards the poles. Classical models of these chemosensory systems are based on the E. coli Che system, which contains only one adaptor protein. We investigated the roles of the two adaptors in Chp array composition and signal transduction. Our initial findings indicated that PilI functions as the primary adaptor in the Chp system and is necessary for signal transduction and array formation to occur, while ChpC is likely an auxiliary protein needed for signal amplification and array stability. Based on the results from bacterial two-hybrid (BACTH) protein-protein interaction studies, we constructed a model for Chp array formation. Using fluorescence microscopy, our newest data on protein localization and array formation further supports this model. The influence of mineral N (MN) on legume-rhizobial symbiosis has been widely studied focusing more on crop legumes that belong to the subfamily Papilionoidea. It has shown that the available MN in soil inhibits or delays nodulation and symbiotic nitrogen fixation. no studies have determined if nodulation in primitive legumes is also responsive to MN. For the determination of the effect of MN on nodulation, in growth The higher SDW of plants with N shows that M. pudica assimilated NO3- and NH4+ but still formed nodules. These results considered together, indicate that nodulation of M. pudica by P. phymatum is likely independent of N availability.

Metabolic Fingerprints of Pitcher Plant Microbiomes Show Dynamic Community Succession

Phosphorus (P), one of the most important macronutrients for plants, is given to the soil through phosphatic fertilizers and organic manures. The application of PSB to soils is one of the most effective and successful ways, since it can replace or reduce the demand for inorganic P fertilizers and could be a long-term strategy for conserving P resources. The goal of the study was to determine the contribution of PSB to phosphorus (P) availability in organic, mineral, and natural sources. For this purpose, a laboratory incubation experiment was conducted at the University of Agriculture Peshawar. The experiment was consisting of four sources of P [poultry manure (PM), farm yard manure (FYM), rock phosphate (RP) and single supper phosphate (SSP)] for obtaining 45 mg P 2 O 5 kg -1 and two types of inoculation (control and PSB) arranged in two factorial complete randomized design with three replications. Resultantly, PSB inoculation significantly improved. P availability at all incubation intervals except day zero. A 3.4%, 5.4%, 12% and 16 % increased were observed in P as compared to control, at day 7, 14, 28 and 56 respectively. Similarly, significant differences were observed in P release for P sources at all incubation intervals. Initially higher P release was observed for SSP but at day 14 onward PM and FYM proved to be the best P releasable sources as compared to mineral P sources. RP released the lowest P at all incubation intervals however it performed statistically at par to SSP at 56 days. The significant interactive effect of P sources and PSB at day 28 and 56 demonstrated that soil receiving P from organic sources inoculated with PSB released higher P than other treatments combination. Thus, it is suggested that P must be applied from organic P sources (PM and FYM) along with PSB inoculation for better P nutrition and crop yield
