Innovative horizons in cancer therapy, imaging, and sensing with Janus Nanoparticles: a comprehensive review

We optimized the loading and release processes of Ciprofloxacin (CIP) on Fe3O4/Chitosan (FCS) magnetic nanoparticles (MNPs) for drug delivery applications. The Fe3O4 MNPs were synthesized via the coprecipitation method and subsequently coated with Chitosan to enhance their properties. Ciprofloxacin was used as a model drug. We characterized the structure, morphology, physicochemical, and magnetic properties of both the Fe3O4/CS MNPs and the CIP-loaded Fe3O4/CS MNPs using various techniques, including SEM, EDX, FTIR, XRD, VSM, TGA, and elemental mapping. In the spectrum of Fe3O4/CS/CIP, the appearance of absorption bands at 3437 cm-1 and 2908 cm-1 for -OH, and -NH2 functional groups of chitosan, the bands at 1378 cm-1 and 1066 cm-1 belong to its CH-OH and -C-O-C groups, and the bands at 1945 cm-1 and 1606 cm-1 of CIP carbonyl groups confirm modification of magnetite NPs by CS and loading CIP by the modified adsorbent. The average crystallite size was obtained about 20 nm based on the diffraction data. The modified adsorbent has a pHpzc of 6.1. The effects of different factors such as pH, time, temperature, and initial concentration on CIP's loading and release processes were studied using response surface methodology (RSM). Our findings indicated that the concentration of CIP was the most significant factor influencing the loading process, while time was the most crucial factor for the release process. The optimal conditions for loading were determined to be at pH 9 with a stirring time of 13 h, whereas the optimal conditions for release were at pH 2.7 with a stirring time of 9 h within a range of 1 to 9 h. One-factor-at-a-time studies indicated that a concentration of 20 ppm and an exposure time of 8 h had the most significant effects on loading and release, respectively.

A chitosan-based magnetic system for response surface methodology (RSM) optimization of the influencing variables in ciprofloxacin loading/releasing

As a packaging materials candidate, based on a polylactic acid/ polybutylene adipate terephthalate (PLA/PBAT) blends (90/10 and 75/25 wt/wt) containing 1, 3, and 5 phr hydrophilic (HPL) and hydrophobic (HPB) nanosilica (NS) particles in the presence of a multifunctional epoxide compatibilizer were prepared by melt mixing, in a twin‐screw extruder. Scanning electron microscopic studies confirmed a matrix‐droplet morphology with finer dispersed domains at higher NS content. Energy dispersive spectroscopy mapping also indicated uniform dispersion of NS in blends, with some agglomerates at higher content of nanoparticles. Moreover, transmission electron microscope was applied to study the impact of nanofillers' localization on the systems' morphology. It was observed that NS particles localized at the PLA‐PBAT interface. In addition, thermogravimetric analysis (TGA) was used to investigate thermal stability and thermal degradation kinetics. Data indicates that hydrophobic NS improved thermal stability. The activation energy of degradation was calculated using several techniques of data modeling, including Friedman, Flynn‐Ozawa‐Wall, and Kissinger‐Akahira‐Sunose models. Among blend nanocomposites, 75/25 blend containing 5 phr HPB NS had the maximum degradation activation energy, suggesting that this sample had the most resistance to heat degradation. The intensity of the TGA/FTIR peaks of the evolved products was found to be correlated with the activation energy. The Criado's technique was also used to investigate the changes in the thermal degradation mechanism.

The advent of Janus nanoparticles has been a great breakthrough in the emerging field of nanomaterials. Janus nanoparticles refer to a single structure with two distinct chemical functions on either side. Owing to their asymmetric structures, they can be utilized in a variety of applications where monomorphic particles are insufficient. In the last decade, a wide variety of materials have been employed to fabricate Janus nanoparticles, and due to the great advantages of magnetite (Iron-oxide) NPs, they have been considered as one of the best candidates. With the main benefit of magnetic controlling, magnetite Janus nanoparticles fulfill great promises, especially in biomedical areas such as bioimaging, cancer therapies, theranostics, and biosensing. The intrinsic characteristics of magnetite Janus nanoparticles (MJNPs) even hold great potential in magnetite Janus forms of micro-/nanomotors. Despite the great interest and potential in magnetic Janus NPs, the need for a comprehensive review on MJNPs with a concentration on magnetite NPs has been overlooked. Herein, we present recent advancements in the magnetite-based Janus nanoparticles in the flourishing field of biomedicine. First, the synthesis and fabrication methods of Janus nanoparticles are discussed. Then we will delve into their intriguing biomedical applications, with a separate section for magnetite Janus micro-/nanomotors in biomedicine. And finally, the challenges and future outlook are provided. Magnetite-based Janus nanoparticles, their synthesis and biomedical applications.

Amylose is a linear polysaccharide with a unique ability to form helical inclusion complexes with the appropriate guest components. Numerous studies have been conducted on encapsulation of bioactive compounds for various applications. In the biomedical field, biohybrid micro/nanomotors (MNMs) have emerged as innovative candidates due to their excellent biocompatible and biodegradable properties. This study was inspired by the biohybrid- and enzymatic-propelled MNMs and explored the potential of amylose inclusion complexes (ICs) in creating these MNMs. The study developed a new type of micromotor made from (PEG-co-PBA)-b-amylose. Nanoprecipitation, dimethyl sulfoxide (DMSO), and ultrasound-treated methods were employed to create spherical, thick crystalline, and rod-bacterial-like morphologies, respectively. Candida antarctica lipase B (CALB) was used as the catalytic fuel to induce the motion by the enzymatic degradation of ester linkages in the polymeric segment. Optical microscopy was utilized to observe the motion of the motors following incubation with enzyme concentrations of 5, 10, and 20% (w/w). The results demonstrated that the velocity of the motors increased proportionally with the percentage of added enzyme. Additionally, a comprehensive molecular docking evaluation with PyRx software provided insight into the interaction of the CALB enzyme with polymeric moieties and demonstrated a good affinity between the enzyme and polymer in the binding site. This study provides novel insight into the design and development of enzymatically driven polymeric micromotors and nanomotors.
