Plant-soil feedback (PSF) may influence plant-insect interactions. Although plant defense differs between shoot and root tissues, few studies have examined root-feeding insect herbivores in a PSF context. We examined here how plant growth and resistance against root-feeding Delia radicum larvae was influenced by PSF. We conditioned soil with cabbage plants that were infested with herbivores that affect D. radicum through plant-mediated effects: leaf-feeding Plutella xylostella caterpillars and Brevicoryne brassicae aphids, root-feeding D. radicum larvae, and/or added rhizobacterium Pseudomonas simiae WCS417r. We analyzed the rhizosphere microbial community, and in a second set of conspecific plants exposed to conditioned soil, we assessed growth, expression of defense-related genes, and D. radicum performance. The rhizosphere microbiome differed mainly between shoot and root herbivory treatments. Addition of Pseudomonas simiae did not influence rhizosphere microbiome composition. Plant shoot biomass, gene expression, and plant resistance against D. radicum larvae was affected by PSF in a treatment-specific manner. Soil conditioning overall reduced plant shoot biomass, Pseudomonas simiae-amended soil causing the largest growth reduction. In conclusion, shoot and root insect herbivores alter the rhizosphere microbiome differently, with consequences for growth and resistance of plants subsequently exposed to conditioned soil.

Pathogen infections seriously threaten plant health and global crop production. Epigenetic processes such as DNA methylation, histone post-translational modifications, chromatin assembly and remodeling play important roles in transcriptional regulation of plant defense responses and could provide a new direction to drive breeding strategies for crop disease resistance improvement. Although past decades have seen unprecedented proceedings in understanding the epigenetic mechanism of plant defense response, most of these advances were derived from studies in model plants like Arabidopsis. In this review, we highlighted the recent epigenetic studies on crop-pathogen interactions and discussed the potentials, challenges, and strategies in exploiting epigenetic variations for crop disease resistance improvement.

Climate change is altering the dynamics of crop pests and diseases resulting in reduced crop yields. Using beneficial soil bacterial to increase crop health is a quickly developing area in sustainable agriculture, but it is unknown if climate change or interactions with other species could alter their effect. The plant growth-promoting rhizobacterium Acidovorax radicis N35 is known to increase barley (Hordeum vulgare) plant growth under laboratory conditions, and we tested the stability of the plant-bacterial interactions when exposed to elevated carbon dioxide (CO2) and ozone (O3) levels while infesting the aboveground leaves with cereal aphids (Sitobion avenae) and the soil with beneficial earthworms. Acidovorax radicis N35 increased plant growth and reduced insect growth - with greatest effect in a high-stress elevated O3 environment, but reduced effects under elevated CO2. Earthworms promoted both plant and insect growth, but inoculation with A. radicis N35 alleviated some of the earthworm-mediated increase in pest abundance, particularly in the ambient environment. The consistency of these beneficial effects highlights the potential of exploiting local species interactions for predicting and mitigating climate change effects in managed systems. We conclude that microbial bioprotectants have high potential for benefiting agriculture via plant-growth promotion and pest suppression.

Microbial-Mediated Plant Growth Promotion and Pest Suppression Varies Under Climate Change

Increasing levels of CO2 and tropospheric ozone (O3) due to climate change are contributing to reduced plant health and unstable crop yield production. The inoculation of plant roots with beneficial fungi or bacteria can increase plant health. However, this is often studied under very controlled conditions and it is unknown how climate change or interactions with other species can alter the resulting benefits. Here we show that the rhizosphere bacterium Acidovorax radicis N35 can increase plant growth and reduce insect growth — with increased impact in a high-stress elevated O3 environment, but reduced impact under elevated CO2. In a fully-factorial climate chamber experiment we disentangled the impacts of climate factors (elevated CO2 and elevated O3) and biotic interactions (plant cultivar, sap-feeding insects and earthworms) on cereal growth and insect suppression mediated by A. radicis N35. Earthworms promoted plant aboveground growth, whereas A. radicis N35 promoted root growth, and overall plant growth was higher when both species were present. However, earthworms also promoted insect growth and therefore increased plant damage through herbivory. While A. radicis N35 inoculation was able to mitigate these negative effects to some extent under an ambient environment this was lost under climate change conditions. Our results show that knowledge-based solutions for sustainable agriculture should include biotic interactions and must be tested across variable climate change scenarios in order to build resilient cropping systems.
