The use of hydrogen peroxide (produced in situ or ex situ) as the main agent in oxidative processes of environmental pollutant removal is widely studied. The degradation of water pollutants, such as dyes, pharmaceuticals, cosmetics, petroleum derivatives, and even pathogens, has been successfully obtained by different techniques. This review gives an overview of the more recent methods developed to apply oxidative processes mediated by H2O2 and other reactive oxygen species (ROS) in environmental catalysis, with particular attention to the strategies (Fenton-like and Bio-Fenton, photo- and electro-catalysis) and the materials employed. A wide discussion about the characteristics of the materials specifically studied for hydrogen peroxide activation, as well as about their chemical composition and morphology, was carried out. Moreover, recent interesting methods for the generation and use of hydrogen peroxide by enzymes were also presented and their efficiency and applicability compared with the Fenton and electro-Fenton methods discussed above. The use of Bio-Fenton and bi-enzymatic methods for the in situ generation of ROS seems to be attractive and scalable, although not yet applied in full-scale plants. A critical discussion about the feasibility, criticalities, and perspectives of all the methods considered completes this review.

An Overview of Environmental Catalysis Mediated by Hydrogen Peroxide

Oxidoreductase (OXR) enzymes are in high demand for biocatalytic applications in food industry and cosmetics: glucose oxidase (GOx), cellobiose dehydrogenase (CBDH), bioremediation: horseradish peroxidase (HRP), laccase (LAC), and medicine for biosensors and miniature biofuel cells (GOx, CBDH, LAC, HRP). They can be used in a soluble form and/or within the yeast cell walls expressed as chimeras on the surface of yeast cells (YSD), such as P. pastoris and S. cerevisiae. However, most of the current studies suffer from either low yield for soluble enzyme expression or low enzyme activity when expressed as chimeric proteins using YSD. This is always the case in studies dealing with heterologous expression of oxidoreductase enzymes, since there is requirement not only for multiple OXR genes integrations into the yeast genome (super transformations), and codon optimization, but also very careful design of fermentation media composition, and fermentation conditions during expression due to need for adding transition metals (copper, iron), and metabolic precursors of FAD and heme. Therefore, scientists are still trying to find optimal formula using above mentioned approaches, and most recently also using protein engineering and directed evolution for additional increase in the yield of recombinant enzyme production. In this review article we will cover all the current state of the art technologies and most recent advances in the field that yielded high expression level for some of these enzymes in specially designed expression/fermentation systems. We will also tackle and discuss new possibilities for further increase in fermentation yield by the cutting-edge technologies such are directed evolution, protein and strain engineering, high-throughput screening methods based on in vitro compartmentalization, flow cytometry and microfluidics. 

State of the Art Technologies for High Yield Heterologous Expression and Production of Oxidoreductase Enzymes: Glucose Oxidase, Cellobiose Dehydrogenase, Horseradish Peroxidase, and Laccases in Yeasts P. pastoris and S. cerevisiae

Glucose oxidase (GOX) catalyzes the FAD-dependent oxidation of α-D-glucose to D-gluconolactone with production of hydrogen peroxide. This enzyme encounters many biotechnological applications from glucose sensors to applications in food, pharma and textile industries. For this purpose, recombinant GOX versions, usually derived from Aspergillus niger, are produced in fermentation systems, frequently in filamentous fungi because other production platforms such as bacteria or yeast have rendered meager results. We wondered whether A. niger GOX, more specifically a mutant version with superior thermotolerant properties, could be efficiently produced in Nicotiana benthamiana plants. To this aim, we used a tobacco mosaic virus-derived vector that is inoculated into plant tissues using A. tumefaciens. Results exhibited the efficient production of the recombinant GOX in plants and the facile downstream purification when the recombinant protein is targeted to the apoplast, the space between plasma membranes and cell walls. The plant-made recombinant GOX displayed excellent catalytic properties in broad pH and temperature conditions. In addition to establishing a new strategy to produce recombinant GOX in plants as a green alternative to traditional fungal fermentation, we further investigated the potential application of this protein as an ezybiotic. Results exhibited a remarkable bacteriocide activity against Escherichia coli and Staphylococcus aureus. 

Production in Nicotiana benthamiana of a thermotolerant glucose oxidase that shows enzybiotic activity against Escherichia coli and Staphylococcus aureus

Here, we developed magnetically recoverable biocatalysts based on magnetite nanoparticles coated with an ultra-thin layer (about 0.9 nm) of chitosan (CS) ionically cross-linked by sodium tripolyphosphate (TPP). Excessive CS amounts were removed by multiple washings combined with magnetic separation. Glucose oxidase (GOx) was attached to the magnetic support via the interaction with N-hydroxysuccinimide (NHS) in the presence of carbodiimide (EDC) leading to a covalent amide bond. These steps result in the formation of the biocatalyst for D-glucose oxidation to D-gluconic acid to be used in the preparation of pharmaceuticals due to the benign character of the biocatalyst components. To choose the catalyst with the best catalytic performance, the amounts of CS, TPP, NHS, EDC, and GOx were varied. The optimal biocatalyst allowed for 100% relative catalytic activity. The immobilization of GOx and the magnetic character of the support prevents GOx and biocatalyst loss and allows for repeated use.

Glucose oxidase, which uses molecular oxygen as an electron acceptor to specifically catalyze the conversion of β-d-glucose to gluconic acid and hydrogen peroxide (H2O2), has been considered an important enzyme in increasing environmental sustainability and food security. However, achieving the high yield, low price and high activity required for commercial viability remains challenging. In this review, we first present a brief introduction, looking at the sources, characteristics, catalytic process, and applications of glucose oxidase. Then, the predictive structures of glucose oxidase from two different sources are comparatively discussed. We summarize the inhibitors of glucose oxidase. Finally, we highlight how the production of glucose oxidase can be improved by optimizing the culture conditions and microbial metabolic engineering.

Insights into the Structures, Inhibitors, and Improvement Strategies of Glucose Oxidase

The preparation of Escherichia coli competent cells by calcium chloride is a common method in molecular biology, but the mechanism is poorly understood. In a previous study, using transcriptomics and proteomics approaches, we found that the expression pattern of the gene loiP was upregulated by CaCl2. In order to investigate the function of the loiP gene in Ca2+ mediated formation of competent cells of E. coli DH5α, the loiP gene deletion strains were constructed by the lambda-derived Red homologous recombination technique. Then the cell morphology, transformation efficiency and cell membrane changes of the competent cells of the mutant strain were further explored. Compared with the wild-type E. coli DH5α, the mutant strains have no significant differences in the morphology, growth characteristics and the permeability of the intracellular membrane. However, the transformation efficiencies of the mutant strains to plasmids of different sizes significantly reduced, and the permeability of the outer membrane decreased by 2.94 times. These results indicate that the deletion of gene loiP may directly affect the transformation efficiency and outer membrane permeability of E. coli competent cells.
