TOX reinforces the phenotype and longevity of exhausted T cells in chronic viral infection.
Francesca Alfei,Kristiyan Kanev,Maike Hofmann,Ming Wu,Hazem E. Ghoneim,Hazem E. Ghoneim,Patrick Roelli,Patrick Roelli,Patrick Roelli,Daniel T. Utzschneider,Madlaina von Hoesslin,Jolie G. Cullen,Yiping Fan,Vasyl Eisenberg,Dirk Wohlleber,Katja Steiger,Doron Merkler,Mauro Delorenzi,Mauro Delorenzi,Percy A. Knolle,Cyrille J. Cohen,Robert Thimme,Benjamin Youngblood,Dietmar Zehn +23 more
TL;DR: It is shown that TOX is a critical factor for the normal progression of T cell dysfunction and the maintenance of exhausted T cells during chronic infection, and provide a link between the suppression of effector function intrinsic to CD8 T cells and protection against immunopathology.

Proliferation-competent Tcf1+ CD8 T cells in dysfunctional populations are CD4 T cell help independent
Kristiyan Kanev,Ming Wu,Antar Drews,Patrick Roelli,Patrick Roelli,Christine Wurmser,Madlaina von Hösslin,Dietmar Zehn +7 more
TL;DR: It is revealed that proliferation-competent cells in dysfunctional populations show a previously unrecognized uncoupling of CD4 T cell help that is otherwise required by conventional memory T cells.

Tailoring the resolution of single-cell RNA sequencing for primary cytotoxic T cells
Kristiyan Kanev,Patrick Roelli,Patrick Roelli,Ming Wu,Christine Wurmser,Mauro Delorenzi,Michael W. Pfaffl,Dietmar Zehn +7 more
TL;DR: In this article, a droplet-based approach for high-throughput analysis (tDrop-seq) and a plate-based method for highperformance in-depth single-cell RNA sequencing (tSCRB-seq).
Pyrimidine de novo synthesis inhibition selectively blocks effector but not memory T cell development
Stefan Scherer,Susanne Oberle,Kristiyan Kanev,Ann-Katrin Gerullis,Ming Wu,Gustavo P. de Almeida,Daniel J. Puleston,Francesc Baixauli,Lilian Aly,A. Greco,T. Nizharadze,Nils B. Becker,Madlaina V Hoesslin,Lara V Donhauser,Jacqueline Berner,Talyn Chu,Hayley A. McNamara,Zeynep Esencan,Patrick Roelli,Christine Wurmser,Ingo Kleiter,Maria J G T Vehreschild,Christoph Mayer,Percy A. Knolle,Martin Klingenspor,Valeria Fumagalli,Matteo Iannacone,Martin Prlic,Thomas Korn,Erika L. Pearce,Thomas Höfer,Anna Maria Schulz,Dietmar Zehn +32 more
TL;DR: This paper showed that memory T cell precursors are resistant to pyrimidine starvation, which is a drug-targetable checkpoint that efficiently diminishes effector T cells without affecting the memory compartment.

PySCNet: A tool for reconstructing and analyzing gene regulatory network from single-cell RNA-Seq data
TL;DR: Wu et al. as discussed by the authors presented PySCNet, a toolkit that enables reconstructing and analyzing gene regulatory networks (GRNs) from single cell transcriptomic data, integrating competitive gene regulatory construction methodologies for cell specific or trajectory specific GRNs and allowing for gene co-expression module detection and gene importance evaluation.
