A Mendelian randomization study of IL6 signaling in cardiovascular diseases, immune-related disorders and longevity.
Mickael Rosa,Arnaud Chignon,Zhonglin Li,Marie-Chloé Boulanger,Benoit J. Arsenault,Yohan Bossé,Sébastien Thériault,Patrick Mathieu +7 more
TL;DR: Gene-based association analysis with S-PrediXcan by using tissues from GTExV7 showed that IL6R tissue expression-disease pair associations were consistent with the directional effect of IL6 signaling identified in MR, which lowers the risk of multiple CVDs and is associated with increased longevity, but at the expense of higher atopic risk.
A transcriptome-wide association study identifies PALMD as a susceptibility gene for calcific aortic valve stenosis.
Sébastien Thériault,Nathalie Gaudreault,Maxime Lamontagne,Mickael Rosa,Marie-Chloé Boulanger,David Messika-Zeitoun,Marie-Annick Clavel,Romain Capoulade,François Dagenais,Philippe Pibarot,Patrick Mathieu,Yohan Bossé +11 more
TL;DR: The identification of PALMD as a susceptibility gene for CAVS provides insights into the genetic nature of this disease, opens avenues to investigate its etiology and to develop much-needed therapeutic options.
DNA methylation of a PLPP3 MIR transposon-based enhancer promotes an osteogenic programme in calcific aortic valve disease.
Ghada Mkannez,Valérie Gagné-Ouellet,Mohamed Jalloul Nsaibia,Marie-Chloé Boulanger,Mickael Rosa,Deborah Argaud,Fayez Hadji,Nathalie Gaudreault,Gabrielle Rhéaume,Luigi Bouchard,Yohan Bossé,Patrick Mathieu +11 more
TL;DR: It is found that PLPP3 gene and enzymatic activity were downregulated in mineralized AVs and DNA methylation of a MIR-based enhancer downregulates the expression of PLPP 3 and promotes the mineralization of the AV.

Enhancer-associated aortic valve stenosis risk locus 1p21.2 alters NFATC2 binding site and promotes fibrogenesis
Arnaud Chignon,Mickael Rosa,Marie-Chloé Boulanger,Deborah Argaud,Romain Devillers,Valentin Bon-Baret,Ghada Mkannez,Zhonglin Li,Anne Rufiange,Nathalie Gaudreault,David Gosselin,Sébastien Thériault,Yohan Bossé,Patrick Mathieu +13 more
TL;DR: In this article, the authors used 3D genetic mapping and CRISPR activation to identify rs6702619 as being located in a distant-acting enhancer, which controls the expression of PALMD.
Age, Sex, and Valve Phenotype Differences in Fibro-Calcific Remodeling of Calcified Aortic Valve.
Martine Voisine,Maxime Hervault,Mylène Shen,Anne-Julie Boilard,Benoit Filion,Mickael Rosa,Yohan Bossé,Patrick Mathieu,Nancy Côté,Marie-Annick Clavel +9 more
TL;DR: Women had less calcification and more fibrotic remodeling than men, regardless of the phenotype of the valve or age of the patient, in calcific aortic valve disease.
