Erlotinib, Gefitinib, and Vandetanib Inhibit Human Nucleoside Transporters and Protect Cancer Cells from Gemcitabine Cytotoxicity
Vijaya L. Damaraju,Tara Scriver,Delores Mowles,Michelle Kuzma,Anderson J. Ryan,Carol E. Cass,Michael B. Sawyer +6 more
TL;DR: The potential for reduced accumulation of nucleoside chemotherapy drugs in tumor tissues due to inhibition of hENTs and/or hCNTs by TKIs indicates that pharmacokinetic properties of these agents must be considered when scheduling TKI and nucleosid chemotherapy in combination.

Multitargeted kinase inhibitors imatinib, sorafenib and sunitinib perturb energy metabolism and cause cytotoxicity to cultured C2C12 skeletal muscle derived myotubes.
TL;DR: Changes in mitochondrial complex activities, glucose and nucleoside uptake leading to decreased energy production and mitochondrial function in a skeletal muscle cell model are caused, suggesting that these changes may play a role in fatigue, one of the most common adverse effects of TKIs.
30
Interactions of Multitargeted Kinase Inhibitors and Nucleoside Drugs: Achilles Heel of Combination Therapy?
TL;DR: In clinical settings, TKI inhibitor concentrations in tumor tissues are sufficient to inhibit hENT1 activity, thereby reducing nucleoside chemotherapy drug levels in cancer cells and reducing efficacy in combination schedules.

Human concentrative nucleoside transporter 3 is a determinant of fludarabine transportability and cytotoxicity in human renal proximal tubule cell cultures
Adam N. Elwi,Adam N. Elwi,Vijaya L. Damaraju,Vijaya L. Damaraju,Michelle Kuzma,Michelle Kuzma,Stephen A. Baldwin,James D. Young,Michael B. Sawyer,Michael B. Sawyer,Carol E. Cass,Carol E. Cass +11 more
TL;DR: Variations in hCNT3 abundance in renal proximal tubules, and hence nucleoside reabsorption, may explain interpatient variability in fludarabine’s pharmacokinetics and toxicities.

Cytotoxic activities of nucleoside and nucleobase analog drugs in malignant mesothelioma: characterization of a novel nucleobase transport activity.
Deepti Damaraju,Vijaya L. Damaraju,Vijaya L. Damaraju,Miranda Brun,Delores Mowles,Delores Mowles,Michelle Kuzma,Michelle Kuzma,Richard Berendt,Richard Berendt,Michael B. Sawyer,Michael B. Sawyer,Carol E. Cass,Carol E. Cass +13 more
TL;DR: Differences in nucleoside analog drug transport activities are likely to contribute to the observed clinical variation in nucleOSide analog response in patients and for the first time a correlation between nucleobase drug sensitivities and transport activities was shown.
