BACKGROUND: Identification of target genes responsible for visible phenotypic effect may contribute to the development of transgene-free bioengineering strategies and application of crop varieties with edited genome. CAPRICE (CPC) is a single-repeat R3 MYB transcription factor, involved in anthocyanin biosynthesis and trichome formation. It is assumed that CPC controls the expression of Dihydroflavonol-4-reductase (DFR), a key gene of anthocyanin biosynthesis. 
AIM: The aim of the study was to determine whether knockout of the CPC gene using CRISPR/Cas9 results in visible anthocyanin accumulation. 
MATERIALS AND METHODS: Three guide RNAs were designed to excise a MYB domain from the CPC gene of Arabidopsis thaliana. Anthocyanin content and expression of CPC and DFR genes were studied in edited plants. 
RESULTS: The expected 662 bp deletion was detected in 2,7% of glufosinate-resistant plants, however none of the mutations were homozygous. Four edited lines were studied in four generations. An upregulation of the DFR gene was observed in edited lines, however CPC gene expression, anthocyanin content and trichome development were not significantly different from those in control plants. Moreover, in A. thaliana pigmentation did not directly depend on DFR or CPC gene expression. 
CONCLUSIONS: Our results suggest that CPC gene is involved in regulation of DFR gene expression and anthocyanin biosynthesis pathway, however in case of mutations plants might utilize other transcription factors to maintain homeostasis. Therefore, CPC gene is not a suitable target for CRISPR/Cas studies in Arabidopsis.

The monograph details and systematises the results of scientific research, including the authors' own research, on the prevalence, harmfulness, biology, development cycle and pathogenesis of major diseases of cruciferous crops. The factors that contribute to the formation of epiphytoties of cruciferous crops diseases from the standpoint of hydrothermal, soil and agrotechnological factors are formulated and described in detail, taking into account the peculiarities of phenostage and physiological development of different types of cruciferous crops. The history of the study of each pathogen and different views on its etiophysiology in conjunction with the biological parameters of growth and development of the main cruciferous species are described in detail. The basic components of the complex of measures to regulate the prevalence and harmfulness of each pathogen separately and in their integrated combination in agrocenoses of cruciferous crops are highlighted, taking into account the world and national experience based on the generalisation of a number of scientific studies over a eighty-year period. The authors of the monograph aimed to summarise the development, prevalence and harmfulness of major cruciferous diseases in Ukraine and the world from the perspective of the results of studies of this issue by scientists from different countries and to summarise the recommendations of Ukrainian and world science (including the results of their own research) on the fundamental and effective technologies for their control in agrocenoses of major cruciferous species, covering a significant period of research on this issue.

Transcription factor caprice as a target to induce anthocyanin biosynthesis in oilseed rape

Oilseed rape (Brassica napus) is an economically important crop. In temperate climate powdery mildew Erysiphe crucifertaum can reduce its yields dra0matically. Nevertheless, cultivars resistant to this fungal disease have not been selected yet. Plants, grown in the field and in the light chamber, were infected with powdery mildew. After 30 days there were severe signs of infection on most of the plants, however several individuals were less susceptible to it. Leaves of B. napus were used to measure the level of GSH, GSSG and GST by spectrophotometric assay. The level of total glutathione was higher in plants with increased resistant to powdery mildew than in both severely infected plants and control plants that were not subjected to the infection. The GST activity in resistant and control plants was at the same level, however in susceptible plants it was lower. Therefore, oilseed rape with naturally increased glutathione content can be used in the selection of resistant cultivars. GST and GSH genes should be also considered as targets for genetic engineering and genome editing.
