The 1980s was a period of intensive research on the N-methyl-D-aspartate (NMDA) receptor. The decade began with the definition of the NMDA receptor as a pharmacologically distinct type of excitatory amino acid (glutamate) receptor (see Watkins i 991), and culminated with a profound understanding of the physiology and biophysics of the receptor, its role in synaptic integration and plasticity, and its potential importance in a surprisingly diverse group of neurological disorders. The 1990s have begun with the cloning of an NMDA receptor gene by Nakanishi and his collaborators (Meguro et al. 1992; Monyer et al. 1992; Moriyoshi et al. 1991). This landmark event will undoubtedly lead to an explosion of information on the structure of the receptor that should initiate a new era in pharmacological targeting of the receptor for neurological therapeutics.
