Efficient production of bispecific IgG of different isotypes and species of origin in single mammalian cells
Michael Dillon,Yiyuan Yin,Jianhui Zhou,Luke McCarty,Diego Ellerman,Dionysos Slaga,Teemu T. Junttila,Guanghui Han,Wendy Sandoval,Meric A. Ovacik,Kedan Lin,Zhilan Hu,Amy Shen,Jacob E. Corn,Christoph Spiess,Paul Carter +15 more
TL;DR: Novel designs to facilitate selective Fab arm assembly in conjunction with previously described knobs-into-holes mutations for preferential heavy chain heterodimerization are described to facilitate single-cell bispecific IgG production.

Development of a Human IgG4 Bispecific Antibody for Dual Targeting of Interleukin-4 (IL-4) and Interleukin-13 (IL-13) Cytokines *
Christoph Spiess,Jack Bevers,Janet Jackman,Nan Chiang,Gerald R. Nakamura,Michael Dillon,Hongfang Liu,Patricia Molina,J. Michael Elliott,Whitney Shatz,Justin Scheer,Glen Giese,Josefine Persson,Yin Zhang,Mark S. Dennis,James Giulianotti,Prateek Gupta,Dorothea Reilly,Enzo Palma,Jianyong Wang,Eric Stefanich,Heleen Scheerens,Germaine Fuh,Lawren C. Wu +23 more
TL;DR: It is shown that IgG4 bispecific antibodies can be generated in large quantities with equivalent efficiency and quality and have comparable pharmacokinetic properties and lung partitioning, compared with the IgG1 isotype.

Characterization of Chain Pairing Variants of Bispecific IgG Expressed in a Single Host Cell by High-Resolution Native and Denaturing Mass Spectrometry
Luis F. Schachner,Guanghui Han,Michael Dillon,Jianhui Zhou,Luke McCarty,Diego Ellerman,Yiyuan Yin,Christoph Spiess,Jennie R. Lill,Paul Carter,Wendy Sandoval +10 more
TL;DR: A mass spectrometric characterization method was developed using native or denaturing conditions by direct infusion into an Exactive Plus Extended Mass Range Orbitrap instrument and Preferential pairing of the anti-IL-13 light chain to its cognate heavy chain was observed, which may be leveraged to guide the design of a single-cell solution for streamlined production of bispecific IgG.

Precise quantification of mixtures of bispecific IgG produced in single host cells by liquid chromatography-Orbitrap high-resolution mass spectrometry.
Yiyuan Yin,Guanghui Han,Jianhui Zhou,Michael Dillon,Luke McCarty,Lou Gavino,Diego Ellerman,Christoph Spiess,Wendy Sandoval,Paul Carter +9 more
TL;DR: A robust high-throughput method for quantitative analysis of bispecific IgG preparations using novel online liquid chromatography in conjunction with an extended mass range Orbitrap-based high-resolution mass spectrometer is developed.

In vitro affinity maturation of a natural human antibody overcomes a barrier to in vivo affinity maturation.
Bing Li,Ashley E. Fouts,Katharina F. Stengel,Peng Luan,Michael Dillon,Wei-Ching Liang,Becket Feierbach,Robert F. Kelley,Isidro Hötzel +8 more
TL;DR: To improve the neutralization potency of natural human antibody MSL-109 targeting human cytomegalovirus (CMV), phage display can be used to identify and bypass barriers to in vivo affinity maturation of antibodies imposed by glycosylation and codon usage.
