Pruning and fertilization practices plays an important role in coffee plantation, used to maintain soil quality and coffee productivity. However, the impact of pruning and fertilization practices on soil microbial activity under coffee-based agroforestry are poorly understood. The aimed of this study was to analyze the response of soil microbial properties (i.e., soil microbial biomass C (SMBC) and soil respiration rates (SR)) to pruning and fertil - ization management in coffee-based agroforestry in UB (Universitas Brawijaya) Forest. A split-plot design with eight treatments and three replications were used in this experiment. The main-plot factor consisted of two types of pruning (Pruning and Bending), and the sub-plot factor consisted of four types of fertilization (i.e., no fertilizer (NF), 100% chicken manure (MN), 50% chicken manure + 50% NPK-inorganic (MN+NPK), and 100% NPK-inorganic (NPK)). The result showed that there was a significantly different (p<0.05) in the soil microbial biomass C and soil respiration after the application of fertilizer. The addition of chicken manure (MN and MN+NPK treat - ment) could enhance the soil microbial biomass and soil respiration, compared to the NF treatment under different pruning practices. The level of soil pH in MN were highest and significantly different with NPK treatment, showed that the application of chicken manure had a potential to neutralize the soil acidity. Metabolic quotient (qCO 2 ) showed highest in the NF treatment as compared to the other treatments. The soil respiration had positive correla - tion (p<0.05) with SMBC, while SMBC had negative correlation (p<0.01) with qCO 2 .

Microbial Biomass and Soil Respiration Response to Pruning and Fertilization Practices in Coffee-Pine Agroforestry

Characterizing the ecological roles of core soil microbial species in soil carbon metabolism is critically important for enhancing carbon sequestration in agricultural systems; however, no studies to date have determined the effects of core soil microbial taxa on carbon metabolism under various long-term fertilization practices. Here, we collected soil samples from field plots that had been subjected to different fertilization practices for nearly 30 years and examined the long-term effects of fertilization on the preferences of core soil bacterial taxa for different carbon sources. We also examined the relative contribution of core soil bacterial taxa in utilization of different carbon source types in Biolog Eco microplates. Long-term fertilization treatment had a significant effect on soil properties and bacterial community structure. The core taxa were closely related to soil carbon source utilization. The co-occurrence network showed that the major ecological clusters containing core taxa made key contributions to soil carbon source utilization. The organic fertilization increased the abundance of a core cluster with a low weighted average rrn copy number. This ecological cluster was the most important factor affecting soil carbon source utilization even among soil physicochemical factors considered. Our findings indicate that core taxa characterized by oligotrophic bacteria have a major effect on carbon source utilization in Ultisols. 

Effects of core soil microbial taxa on soil carbon source utilization under different long-term fertilization treatments in Ultisol

Microorganisms regulate the responses of terrestrial ecosystems to anthropogenic nutrient inputs. The escalation of anthropogenic activities has resulted in a rise in the primary terrestrial constraining elements, namely nitrogen (N) and phosphorus (P). Nevertheless, the specific mechanisms governing the influence of soil microbial community structure and ecological processes in ecologically vulnerable and delicate semi-arid loess agroecosystems remain inadequately understood. Therefore, we explored the effects of different N and P additions on soil microbial community structure and its associated ecological processes in the farmland of Chinese Loess Plateau based on a 36-a long-term experiment. Nine fertilization treatments with complete interactions of high, medium, and low N and P gradients were set up. Soil physical and chemical properties, along with the microbial community structure were measured in this study. Additionally, relevant ecological processes such as microbial biomass, respiration, N mineralization, and enzyme activity were quantified. To elucidate the relationships between these variables, we examined correlation-mediated processes using statistical techniques, including redundancy analysis (RDA) and structural equation modeling (SEM). The results showed that the addition of N alone had a detrimental effect on soil microbial biomass, mineralized N accumulation, and β-1,4-glucosidase activity. Conversely, the addition of P exhibited an opposing effect, leading to positive influences on these soil parameters. The interactive addition of N and P significantly changed the microbial community structure, increasing microbial activity (microbial biomass and soil respiration), but decreasing the accumulation of mineralized N. Among them, N24P12 treatment showed the greatest increase in the soil nutrient content and respiration. N12P12 treatment increased the overall enzyme activity and total phospholipid fatty acid (PLFA) content by 70.93%. N and P nutrient contents of the soil dominate the microbial community structure and the corresponding changes in hydrolytic enzymes. Soil microbial biomass, respiration, and overall enzyme activity are driven by mineralized N. Our study provides a theoretical basis for exploring energy conversion processes of soil microbial community and environmental sustainability under long-term N and P additions in semi-arid loess areas.
