Volumetric absorptive microsampling (VAMS) is a novel microsampling technology that is frequently used for the collection of biofluids for the application in a plethora of bioanalytical applications and assays. Recently, few papers have reported the successful application of VAMS in untargeted as well in targeted metabolomics. VAMS in metabolomics can become in the near future a very promising microsampling technique, but to that moment, some application-related aspects of VAMS in the metabolomics workflow still needs some in depth investigation and evaluation. In this mini-review article, the application of VAMS in metabolomics, procedures applied and first results obtained are described, evaluated and discussed.

Biofluid Collection in Metabolomics by the Application of the novel Volumetric Absorptive Microsampling Technology: a mini-Review

Objective: Pharmacokinetic evaluation of montelukast sodium chronomodulated capsules (sustained-release solid dispersion of drug enclosed in pH-sensitive film-coated hard gelatin shell) and marketed tablets has been carried out in this study. \nMethods: A single oral dose of prepared capsules and marketed conventional tablets was administered in healthy male Dunkin-Hartley albino guinea pigs. Blood samples were collected at different time intervals and plasma concentration of drug was determined by reversed-phase high-performance liquid chromatography. Different pharmacokinetic parameters were assessed from plasma drug concentration-time profile by one-compartment model, first-order kinetics. \nResults: Pharmacokinetic parameters such as time to reach maximum concentration, elimination rate constant, elimination half-life, and mean residence time data indicates that drug release from chronomodulated capsules is significantly prolonged with initial release lag time of 3.5–4 h in comparison with marketed conventional tablets. However, maximum drug plasma concentration, area under the concentration-time curve, and apparent volume of distribution values show non-significant difference between capsules and marketed tablets. \nConclusion: The findings specified that capsules were providing time controlled delivery of drug at a desired rate for prolonged time, which may be helpful for the prevention of episodic attack of asthma in early morning hours.

The evolution of new variants of plant pathogens is one of the biggest challenges to controlling and managing plant diseases. Of the forces driving these evolutionary processes, global migration events are particularly important for widely distributed diseases such as the potato late blight, caused by the oomycete Phytophthora infestans (Mont.) de Bary. However, little is known about its migration routes outside North America and Europe. This work used genotypic data from population studies to elucidate the migration history originating the Colombian P. infestans population. For this purpose, a dataset of 1706 P. infestans genotypes was recollected, representing North and South America, Europe, and Asia. Descriptive analysis and historical records from North America and Europe were used to propose three global migration hypotheses, differing on the origin of the disease (Mexico or Peru) and the hypothesis that it came back to South America from Europe. These scenarios were tested using Approximate Bayesian Computation. According to this analysis, the most probable scenario (posterior probability = 0.631) was the one proposing a Peruvian origin for P. infestans, an initial migration towards Colombia and Mexico, and a later event from Mexico to the United States, and then to Europe and Asia, with no return to northern South America. In Colombia, the scenario considering a single migration from Peru and posterior migrations within Colombia was the most probable with a posterior probability of 0.640. The obtained results support the hypothesis of a Peruvian origin for P. infestans followed by rare colonization events worldwide.

This paper reports the development and validation of different types of dried plasma spot bioassays and their application in a pharmacokinetic study. The pharmacokinetic study involved a single oral dose of the drug acetaminophen and post-dose blood samples were collected by finger prick. Plasma was separated for dried plasma spot preparation and thereafter quantitatively analyzed by liquid chromatography-accurate mass high resolution mass spectrometry. Comparison of results from bioanalytical performance experiments indicated that the two dried plasma spot bioassays worked well and observed acetaminophen pharmacokinetic values were found not to be statistically significantly different between the evaluated two dried plasma spot bioassays. More over, pharmacokinetic values observed were also not statistically significant different from values reported in literature. Overall, it was concluded that dried plasma spot bioassays could have a great potential as alternative sampling technique in pharmacokinetic studies.
