The RPAP3-Cterminal domain identifies R2TP-like quaternary chaperones.
Chloé Maurizy,Marc Quinternet,Yoann Abel,Céline Verheggen,Paulo E. Santo,Maxime Bourguet,Ana C. F. Paiva,Benoît. Bragantini,Marie-Eve Chagot,Marie-Cécile Robert,Claire Abeza,Philippe Fabre,Philippe Fort,Franck Vandermoere,Pedro M. F. Sousa,Jean-Christophe Rain,Bruno Charpentier,Sarah Cianférani,Tiago M. Bandeiras,Bérengère Pradet-Balade,Xavier Manival,Edouard Bertrand +21 more
TL;DR: A structural approach is used to study RPAP3 and find an RP AP3-like protein (SPAG1) which also forms a co-chaperone complex with PIH1D2 and RUVBL1/2 enriched in testis, indicating that R2SP functions in quaternary protein folding.
Recurrent activating mutations of PPAR gamma associated with luminal bladder tumors.
Natacha Rochel,Clémentine Krucker,L. Coutos-Thevenot,L. Coutos-Thevenot,Judit Osz,R. Zhang,R. Zhang,R. Zhang,E. Guyon,E. Guyon,W. Zita,S. Vanthong,O.A. Hernandez,Maxime Bourguet,K.A. Radvanyi,F. Radvanyi,Isabelle Bernard-Pierrot,Isabelle Bernard-Pierrot +37 more
TL;DR: Genomic alterations of PPARG that lead to pro-tumorigenic PPARγ/RXRα pathway activation in luminal bladder tumors are revealed and may open the way towards alternative options for treatment.

Mechanistic insights into the synergistic activation of the RXR-PXR heterodimer by endocrine disruptor mixtures.
Vanessa Delfosse,Tiphaine Huet,Deborah Harrus,Meritxell Granell,Maxime Bourguet,Caroline Gardia-Parège,Barbara Chiavarina,Marina Grimaldi,Sébastien Le Mével,Pauline Blanc,David Huang,Jakub Gruszczyk,Barbara A. Demeneix,Sarah Cianférani,Jean-Baptiste Fini,Patrick Balaguer,William Bourguet +16 more
TL;DR: In this paper, a set of cell-based, biophysical, structural, and in vivo approaches were combined to identify and characterize additional active mixtures, and reveal previously unidentified binding mechanisms involving dynamic structural transitions and covalent coupling.

Deep Structural Analysis of RPAP3 and PIH1D1, Two Components of the HSP90 Co-chaperone R2TP Complex
Julien Henri,Marie-Eve Chagot,Maxime Bourguet,Yoann Abel,Guillaume Terral,Chloé Maurizy,Christelle Aigueperse,Florian Georgescauld,Franck Vandermoere,Rénette Saint-Fort,Isabelle Behm-Ansmant,Bruno Charpentier,Bérengère Pradet-Balade,Céline Verheggen,Edouard Bertrand,Philippe Meyer,Sarah Cianférani,Xavier Manival,Marc Quinternet +18 more
TL;DR: A deep structural investigation of the HSP binding abilities of the two TPR domains of RPAP3 is performed and the existence of a complex lacking PIH1D1 in human cells (R2T), which shows differential binding to certain clients is confirmed.

Pushing the limits of native MS: Online SEC-native MS for structural biology applications
Evolène Deslignière,Marie Ley,Maxime Bourguet,Anthony Ehkirch,Thomas Botzanowski,Stéphane Erb,Oscar Hernandez-Alba,Sarah Cianférani +7 more
TL;DR: It is shown that online SEC coupling can also serve as the basis for multidimensional non-denaturing liquid chromatography workflows, with the SEC acting as a fast desalting device, helping to achieve first dimension LC separation in optimal chromatographic conditions while being compatible with further nMS analysis.
