Yeast mitochondrial Gln-tRNAGln is generated by a GatFAB-mediated transamidation pathway involving Arc1p-controlled subcellular sorting of cytosolic GluRS
Mathieu Frechin,Bruno Senger,Mélanie Brayé,Daniel Kern,Robert P. Martin,Hubert Dominique Becker +5 more
TL;DR: Yeast mitochondrial glutaminyl-tRNA(Gln) is in fact generated by a transamidation pathway involving a novel type of trimeric tRNA-dependent amidotransferase (AdT), and this novel strategy that enables a single protein to be localized in both the cytosol and mitochondria provides a new paradigm for regulation of the dynamic subcellular distribution of proteins between membrane-separated compartments.
Cell-intrinsic adaptation of lipid composition to local crowding drives social behaviour
Mathieu Frechin,Thomas Stoeger,Stephan Daetwyler,Charlotte Gehin,Nico Battich,Eva-Maria Damm,Lilli Stergiou,Howard Riezman,Lucas Pelkmans +8 more
TL;DR: A cell-intrinsic molecular mechanism that allows multicellular patterning without requiring specific communication between cells is revealed and suggests a fundamental role for a tunable membrane lipid composition in collective cell behaviour.

Expression of nuclear and mitochondrial genes encoding ATP synthase is synchronized by disassembly of a multisynthetase complex.
Mathieu Frechin,Ludovic Enkler,Emmanuel Tetaud,Emmanuel Tetaud,Daphné Laporte,Bruno Senger,Corinne Blancard,Corinne Blancard,Philippe Hammann,Gaétan Bader,Sandra Clauder-Münster,Lars M. Steinmetz,Lars M. Steinmetz,Robert P. Martin,Jean-Paul di Rago,Jean-Paul di Rago,Hubert Dominique Becker +16 more
TL;DR: This work shows that the AME complex coordinates expression of enzymes that require intergenomic control that depends on a cytosolic complex made of two aminoacyl-tRNA synthetases attached to an anchor protein, Arc1p.

Crystal structure of a transfer-ribonucleoprotein particle that promotes asparagine formation
Mickaël Blaise,Marc Bailly,Mathieu Frechin,Manja A. Behrens,Frédéric Fischer,Cristiano L. P. Oliveira,Hubert Dominique Becker,Jan Skov Pedersen,Søren Thirup,Daniel Kern +9 more
TL;DR: It is proposed that the crystal structure of the Thermus thermophilus transamidosome represents a transient state of the transamidation reaction, in which tRNAs serve the function of both substrate and structural foundation for a large molecular machine.
Translating organellar glutamine codons: a case by case scenario?
TL;DR: The pathway for Q-tRNAQ synthesis in yeast and mammals mitochondria is still questionable, but QRS activity was found in protozoan mitochondria while AdT activity was characterized in plant organelles.
