Over the past decade, plant biostimulants have been increasingly used in agriculture as environment-friendly tools that improve the sustainability and resilience of crop production systems under environmental stresses. Protein hydrolysates (PHs) are a main category of biostimulants produced by chemical or enzymatic hydrolysis of proteins from animal or plant sources. Mostly composed of amino acids and peptides, PHs have a beneficial effect on multiple physiological processes, including photosynthetic activity, nutrient assimilation and translocation, and also quality parameters. They also seem to have hormone-like activities. Moreover, PHs enhance tolerance to abiotic stresses, notably through the stimulation of protective processes such as cell antioxidant activity and osmotic adjustment. Knowledge on their mode of action, however, is still piecemeal. The aims of this review are as follows: (i) Giving a comprehensive overview of current findings about the hypothetical mechanisms of action of PHs; (ii) Emphasizing the knowledge gaps that deserve to be urgently addressed with a view to efficiently improve the benefits of biostimulants for different plant crops in the context of climate change.

Biostimulant Properties of Protein Hydrolysates: Recent Advances and Future Challenges

The effects of different types of biostimulants on crops include improving the visual quality of the final products, stimulating the immune systems of plants, inducing the biosynthesis of plant defensive biomolecules, removing heavy metals from contaminated soil, improving crop performance, reducing leaching, improving root development and seed germination, inducing tolerance to abiotic and biotic stressors, promoting crop establishment and increasing nutrient-use efficiency. Protein hydrolysates are mixtures of polypeptides and free amino acids resulting from enzymatic and chemical hydrolysis of agro-industrial protein by-products obtained from animal or plant origins, and they are able to alleviate environmental stress effects, improve growth, and promote crop productivity. Amino acids involve various advantages such as increased yield and yield components, increased nutrient assimilation and stress tolerance, and improved yield components and quality characteristics. They are generally achieved through chemical or enzymatic protein hydrolysis, with significant capabilities to influence the synthesis and activity of some enzymes, gene expression, and redox-homeostasis. Increased yield, yield components, and crop quality; improved and regulated oxidation-reduction process, photosynthesis, and physiological activities; decreased negative effects of toxic components; and improved anti-fungal activities of plants are just some of the more important benefits of the application of phenols and phenolic biostimulants. The aim of this manuscript is to survey the impacts of amino acids, different types of protein hydrolysates, phenols, and phenolic biostimulants on different plants by presenting case studies and successful paradigms in several horticultural and agricultural crops.

Amino Acids Biostimulants and Protein Hydrolysates in Agricultural Sciences

Τhe aim of this study was to examine the potential of using biostimulants for the amelioration of deficit irrigation effects on field-grown lettuce plants growth parameters (cv. Doris (Romaine type) and cv. Manchester (Batavia type)). Therefore, five biostimulatory products that differed in their composition were evaluated, including seaweed extracts, amino acids, humic and fulvic acids, macronutrients, Si, and vegetable proteins, while a control treatment with no biostimulants applied on plants was also considered. Plants were subjected to three irrigation regimes, e.g., rain-fed plants (RF), deficit irrigation (I1; 50% of field capacity) and normal irrigation (I2; 100 of field capacity). The results indicate that the application of seaweed extracts, macronutrients, and amino acids (SW treatment) alleviated the negative effects of deficit irrigation on plant growth and chlorophyll content of Romaine-type plants. On the other hand, Batavia-type plants were more susceptible to water stress, since the highest crop yield plant was observed under the full irrigation treatment and the application of vegetal proteins and amino acids (VP treatment). In general, the application of biostimulants on the Romaine type improved plant growth under water shortage conditions compared with fully irrigated plants in almost all measurements, whereas the Batavia-type plants appeared to be more sensitive to deficit irrigation. Therefore, the ecofriendly practices of deficit irrigation and biostimulant application could be useful in leafy vegetable production on a genotype-depended manner.

The Alleviation Effects of Biostimulants Application on Lettuce Plants Grown under Deficit Irrigation

Ensuring the safety of crop production presents a significant challenge to humanity. Pesticides and fertilizers are commonly used to eliminate external interference and provide nutrients, enabling crops to sustain growth and defense. However, the addition of chemical substances does not meet the environmental standards required for agricultural production. Recently, natural sources such as biostimulants have been found to help plants with growth and defense. The development of biostimulants provides new solutions for agricultural product safety and has become a widely utilized tool in agricultural. The review summarizes the classification of biostimulants, including humic-based biostimulant, protein-based biostimulant, oligosaccharide-based biostimulant, metabolites-based biostimulants, inorganic substance, and microbial inoculant. This review attempts to summarize suitable alternative technology that can address the problems and analyze the current state of biostimulants, summarizes the research mechanisms, and anticipates future technological developments and market trends, which provides comprehensive information for researchers to develop biostimulants. 

Plant Biostimulant as an Environmentally Friendly Alternative to Modern Agriculture.

The availability of irrigation water of good quality is decreasing due to soil salinization and the deterioration of aquifers. Moreover, ongoing climate change severely affects crop production and necessitates the intensification of cropping systems in order to ensure food security at a global scale. For this purpose, the aim of the present study was to evaluate the mitigating effects of two natural biostimulants on Diplotaxis tenuifolia L. plants cultivated at different salinity levels (EC of 0 dS m−1, 2 dS m−1, 4 dS m−1, and 6 dS m−1) and harvested at six consecutive cropping cycles. The tested factors showed a varied combinatorial effect on the tested parameters. These findings indicate the importance of considering growing conditions and cropping periods when applying biostimulants in D. tenuifolia plants under salinity stress. Antioxidant activity and bioactive compounds, such as total phenols, carotenoids, and total ascorbic acid, were variably affected by salinity, biostimulant application, and harvesting time, while mineral profile was also affected by the tested factors depending on the combination of factors. Finally, nitrate content showed decreasing trends with increasing salinity, while biostimulant application resulted in the higher accumulation of nitrates compared to the untreated plants. Although biostimulant application seems to alleviate the negative effects of salinity stress, the effect of growing conditions, as indicated by successive crop cycles, is also important for the response of D. tenuifolia plants to saline conditions and biostimulant application.

Bioactive Compounds, Antioxidant Activity, and Mineral Content of Wild Rocket (Diplotaxis tenuifolia L.) Leaves as Affected by Saline Stress and Biostimulant Application

Water deficit causes substantial yield losses that climate change is going to make even more problematic. Sustainable agricultural practices are increasingly developed to improve plant tolerance to abiotic stresses. One innovative solution amongst others is the integration of plant biostimulants in agriculture. In this work, we investigate for the first time the effects of the biostimulant –Leafamine®–a protein hydrolysate on greenhouse lettuce (Lactuca sativa L.) grown under well-watered and water-deficit conditions. We examined the physiological and metabolomic water deficit responses of lettuce treated with Leafamine® (0.585 g/pot) or not. Root application of Leafamine® increased the shoot fresh biomass of both well-watered (+40%) and deficit-irrigated (+20%) lettuce plants because the projected leaf area increased. Our results also indicate that Leafamine® application could adjust the nitrogen metabolism by enhancing the total nitrogen content, amino acid (proline) contents and the total protein level in lettuce leaves, irrespective of the water condition. Osmolytes such as soluble sugars and polyols, also increased in Leafamine®-treated lettuce. Our findings suggest that the protective effect of Leafamine is a widespread change in plant metabolism and could involve ABA, putrescine and raffinose.
