Bacteriophages are increasingly being used as biological control agents against pathogenic bacteria. In the present study, we isolate and characterize bacteriophage Akh-2 from Geoje Island, South Korea, to evaluate its utility in controlling motile Aeromonas septicemia. Akh-2 lysed four of the seven Aeromonas hydrophila strains tested. Transmission electron microscopy analysis showed that Akh-2 belongs to the Siphoviridae family, with head and tail sizes of 50 ± 5 and 170 ± 5 nm, respectively. One-step growth curve analysis revealed that the phage has a latent period of 50 ± 5 min and a burst size of 139 ± 5 plaque-forming units per infected cell. The phage appeared stable in a pH range of 6-8 and a temperature range of -80 to 46 °C. Based on next-generation sequencing analysis, its genome is 114,901 bp in size, with a 44.22% G + C content and 254 open reading frames. During an artificial induction of the disease, loach (Misgurnus anguillicaudatus) treated with Akh-2 showed an increased survival rate and time compared with the non-treated control. Our results suggest that Akh-2 is a potential biological agent for the treatment of Aeromonas infections in fish.

Isolation, Characterization, and Application of a Bacteriophage Infecting the Fish Pathogen Aeromonas hydrophila.

Inland aquaculture contributed by three major fish groups, including carps, tilapias, and catfish plays a vital role in global food security and nutrition, particularly in low and middle-income countries. However, the sustainable development of this sector is hampered by disease epidemics, especially those caused by bacteria such as Aeromonas species. At least eight pathogenic motile Aeromonas species (A. hydrophila, A. veronii, A. jandaei, A. caviae, A. sobria, A. bestiarum, A. dhakensis and A. schubertii) have been reported in aquaculture with some causing up to 100% mortality during disease outbreaks. Simultaneously, emerging multidrug-resistant Aeromonas due to a long-inappropriate use of antibiotics is alarming and highlights a global public health concern and negative socioeconomic impacts. Here, we provide a comprehensive overview of motile Aeromonas infections, antibiotic use and antimicrobial resistance of Aeromonas species. This contribution also highlights the non-antibiotic approaches (the solutions for preventing or treating of bacterial diseases without resorting to antibiotic use) to control motile Aeromonas infections. In addition to the current state of knowledge and limitations of each prophylaxis/therapy, perspectives for future research are discussed critically, including oral/immersion multivalent vaccines, microencapsulated synbiotics, exogenous metabolites, and novel lytic bacteriophage cocktails. Some emerging applicable nanotechnology themes such as nanovaccines, nanobioactive compounds, and nanobubbles are also included in this review. In summary, combating motile Aeromonas infections in aquaculture, including multidrug-resistant aeromonads, as well as other bacterial diseases, is a lengthy battle that requires a strategic combination of multiple non-antibiotic approaches coherent with the One Health philosophy.

The present study was made to assess the effects of dietary Citrus sinensis peel extract on the growth performance, digestive enzyme activity, muscle biochemical compositions, profiles of fatty acid and amino acid, and metabolic enzyme status of the freshwater fish Catla catla. The methanolic extract of C. sinensis peel was supplemented with basal diets at 2, 6, and 10 g kg−1 and fed to C. catla for a 45-day experiment period. Fish fed with the different concentrations of C. sinensis peel-supplemented extract showed significant (P ˂ 0.05) improvement in the survival, growth, muscle biochemical compositions, digestive enzyme activities, and profile of amino acids and fatty acids when compared to control. Among these different concentrations, 6 g kg− 1 C. sinensis-supplemented diet produced a significantly better performance when compared to other concentrations. Similarly, the insignificant (P > 0.05) difference was observed in the metabolic enzyme activities (glutamic oxaloacetic transaminase and glutamic pyruvic transaminase) in all concentrations of C. sinensis peel-supplemented diet-fed fish. It indicates that the supplemented peel extract did not produce any adverse effect on C. catla. The obtained results suggested that the 6 g kg− 1 of C. sinensis can be supplemented in the diet of C. catla for regulating better survival and growth.

Isolation and characterization of phage AHP-1 and its combined effect with chloramphenicol to control Aeromonas hydrophila.

Phage therapy is an alternative approach to overcome the problem of multidrug-resistant bacteria. Here, a novel bacteriophage AhyVDH1, which infects Aeromonas hydrophila 4572, was isolated and its morphology, one-step growth curve, lytic activity, stability under various conditions, and genome were investigated. Transmission electron microscopy revealed that AhyVDH1 has an icosahedral head 49 nm in diameter and a contractile tail 127 nm in length, suggesting that it belongs to the family Myoviridae. AhyVDH1 showed strong adsorption to the surface of A. hydrophila 4572 (90% in 10 min). The latent period of AhyVDH1 was shown to be 50 min, and the burst size was 274 plaque-forming unit/infected cell. AhyVDH1 was stable at 30 °C for 1 h and lost infectivity after20 min of heating at 60 °C. Infectivity remained unaffected at pH 6–7 for 1 h, while the bacteriophage was inactivated at pH   11. AhyVDH1 has a 39,175-bp genome, with a 58% G + C content and 59 open reading frames. BLAST analysis indicated that the genome sequence of phage AhyVDH1 was related to that of Aeromonas phage Ahp2. Both time and MOI-dependent in vitro A. hydrophila growth inhibition were observed with AhyVDH1.Re-growth of the host bacteria appeared about 12 h after treatment, suggesting its potential therapeutic value in treating A. hydrophila infections, but phage cocktails should be developed.

Characterization of Novel Bacteriophage AhyVDH1 and Its Lytic Activity Against Aeromonas hydrophila

Infections with Aeromonas hydrophila is a progressive problem in aquaculture. The use of antibiotic such as Ciprofloxacine has contributed to the rapid and effective treatment of disease cause by this organism. However the Fast-paced increase of resistance to the Said antibiotics has posed problems and there is now a new approach to look for alternative method to control this bacterial pathogen. Phage therapy comes in as a new method to respond to these growing problems. This study demonstrated the promising action of isolated bacteriophage ΦZH1 and ΦZH2 for therapy against Motile Aeromonas Septicemia in Nile tilapia caused by Aeromonas hydrophila.
