Structure of active β-arrestin-1 bound to a G-protein-coupled receptor phosphopeptide
Arun K. Shukla,Aashish Manglik,Andrew C. Kruse,Kunhong Xiao,Rosana I. Reis,Wei Chou Tseng,Dean P. Staus,Daniel Hilger,Serdar Uysal,Li-Yin Huang,Marcin Paduch,Prachi Tripathi-Shukla,Akiko Koide,Shohei Koide,William I. Weis,Anthony A. Kossiakoff,Brian K. Kobilka,Robert J. Lefkowitz,Robert J. Lefkowitz +18 more
TL;DR: The crystal structure of β-arrestin-1 in complex with a fully phosphorylated 29-amino-acid carboxy-terminal peptide derived from the human V2 vasopressin receptor is reported, and results indicate a potentially general molecular mechanism for activation of these multifunctional signalling and regulatory proteins.
Architecture of the fungal nuclear pore inner ring complex
Tobias Stuwe,Christopher J. Bley,Karsten Thierbach,Stefan Petrovic,Sandra Schilbach,Daniel J. Mayo,Thibaud Perriches,Emily J. Rundlet,Young E. Jeon,Leslie N. Collins,Ferdinand M. Huber,Daniel H. Lin,Marcin Paduch,Akiko Koide,Vincent Lu,Jessica Fischer,Ed Hurt,Shohei Koide,Anthony A. Kossiakoff,André Hoelz +19 more
TL;DR: The reconstitution and interdisciplinary analyses of the ~425-kilodalton inner ring complex (IRC), which forms the central transport channel and diffusion barrier of the NPC, are presented, revealing its interaction network and equimolar stoichiometry.

Assessment of a method to characterize antibody selectivity and specificity for use in immunoprecipitation
Edyta Marcon,Harshika Jain,Anandi Bhattacharya,Hongbo Guo,Sadhna Phanse,Shuye Pu,Gregory Byram,Ben C. Collins,Evan Dowdell,Maria Fenner,Xinghua Guo,Ashley Hutchinson,Jacob J. Wodak,Susanne Gräslund,Jack Greenblatt,Aled M. Edwards +43 more
TL;DR: This paper quantified the abundance of all the proteins in immunoprecipitates of 1,124 new recombinant antibodies for 152 chromatin-related human proteins by comparing normalized spectral abundance factors from the target antigen with those of all other proteins.

Structure of the Get3 targeting factor in complex with its membrane protein cargo
Agnieszka Mateja,Marcin Paduch,Hsin Yang Chang,Anna Szydlowska,Anthony A. Kossiakoff,Ramanujan S. Hegde,Robert J. Keenan +6 more
TL;DR: The physiologic assembly pathway for a functional targeting complex was reconstituted and it was shown that it comprises a TA protein bound to a Get3 homodimer, elucidate the mechanism of TA protein recognition and shielding by Get3 and suggest general principles of hydrophobic domain chaperoning by cellular targeting factors.

Structural basis for activation of SAGA histone acetyltransferase Gcn5 by partner subunit Ada2.
Jian Sun,Marcin Paduch,Sang Ah Kim,Ryan M. Kramer,Adam F. Barrios,Vincent Lu,Judy Luke,Svitlana Usatyuk,Anthony A. Kossiakoff,Song Tan +9 more
TL;DR: Structural and biochemical results indicate that the Ada2 SANT domain does not activate Gcn5’s activity by directly affecting histone peptide binding as previously proposed, and suggests a mechanism for regulating chromatin modification enzyme activity: controlling binding of the modification cosubstrate instead of the histone substrate.
