Carbosilane dendrimers with phosphonium terminal groups are low toxic non-viral transfection vectors for siRNA cell delivery.
Regina Herma,Dominika Wrobel,Michaela Liegertová,Monika Müllerová,Tomáš Strašák,Marek Maly,Alena Semerádtová,Marcel Štofik,Dietmar Appelhans,Jan Maly +9 more
TL;DR: 3rd generation carbosilane dendrimers possessing (trimethyl)phosphonium (PMe3) groups on their periphery were able to effectively deliver the functional siRNA into the cells, release it into the cytosol and achieve up to 40% gene silencing of targeted gene with the comparable or, in some cases, even better effectivity as their ammonium counterparts.

Evaluation of toxicological and teratogenic effects of carbosilane glucose glycodendrimers in zebrafish embryos and model rodent cell lines
Michaela Liegertová,Dominika Wrobel,Regina Herma,Monika Müllerová,Lucie Červenková Šťastná,Petra Cuřínová,Tomáš Strašák,Marek Malý,Jan Čermák,Jiří Smejkal,Marcel Štofik,Jan Maly +11 more
TL;DR: The modified FET test showed a two to three orders of magnitude difference between the in vitro cytotoxicity and in vivo developmental toxicity of DDM3Glu, suggesting that their developmental toxicity should be thoroughly investigated to exclude safety risks associated with their potential biomedical use.

Class II biocompatible E-Shell 300 3D printing material causes severe developmental toxicity in Danio rerio embryos and reduced cell proliferation in vitro – implications for 3D printed microfluidics
Zuzana Nejedlá,David Poustka,Regina Herma,Michaela Liegertová,Marcel Štofik,Jiří Smejkal,Vaclav Sicha,Pavel Kaule,Jan Malý +8 more
TL;DR: The results indicate the importance of biocompatibility testing of the 3D printing photopolymer material in direct contact with the given biological model and the possibility of eliminating toxic effects by an appropriate post-processing strategy opens the door for broader applications of E-Shell 300 photopolymers in the development of complex microfluidic devices for various biological applications.
A simple approach for fabrication of optical affinity-based bioanalytical microsystem on polymeric PEN foils.
Alena Semerádtová,Marcel Štofik,O. Neděla,Ondřej Staněk,Petr Slepička,Zdeňka Kolská,Jan Malý +6 more
TL;DR: It is concluded that the presented novel concept enabled us to micropattern functional biosensing layers on the surface of PEN foil in a fast and easy way for continuous roll-to-roll fabrication of low-cost optical bioanalytical devices.

Biocompatible Size-Defined Dendrimer-Albumin Binding Protein Hybrid Materials as a Versatile Platform for Biomedical Applications.
Jan Maly,Ondrej Stanek,Jan Frolik,Marek Maly,Franka Ennen,Dietmar Appelhans,Alena Semerádtová,Dominika Wrobel,Marcel Štofik,Tereza Knapova,Milan Kuchar,Lucie Červenková Št’astná,Jan Čermák,Peter Sebo,Petr Maly +14 more
TL;DR: The synthesis and the characterization of biohybrid conjugates based on noncovalent conjugation between a multivalent biotinylated and PEGylated poly(amido amine) dendrimer and a tetrameric streptavidin-small protein binding scaffold are reported on.
