Heterozygous Mutations Causing Partial Prohormone Convertase 1 Deficiency Contribute to Human Obesity
John W.M. Creemers,Hélène Choquet,Pieter Stijnen,Vincent Vatin,Marie Pigeyre,Sigri Beckers,Sandra Meulemans,Manuel E. Than,Loic Yengo,Maithé Tauber,Beverley Balkau,Paul Elliott,Marjo-Riitta Järvelin,Wim Van Hul,Luc Van Gaal,Fritz F. Horber,François Pattou,Philippe Froguel,David Meyre +18 more
TL;DR: The results provide the first evidence of an increased risk of obesity in heterozygous carriers of mutations in the PCSK1 gene, which is present in 0.83% of extreme obesity phenotypes.

X-ray Structures of Human Furin in Complex with Competitive Inhibitors.
Sven O. Dahms,Kornelia Hardes,Gero L. Becker,Torsten Steinmetzer,Hans Brandstetter,Manuel E. Than +5 more
TL;DR: This work presents a novel preparation of human furin and the first crystal structures of this enzyme in complex with noncovalent inhibitors, and shows the inhibitor exchange by soaking, allowing the investigation of additional inhibitors and substrate analogues.

Novel Furin Inhibitors with Potent Anti-infectious Activity
Kornelia Hardes,Gero L. Becker,Yinghui Lu,Sven O. Dahms,Susanne M. Köhler,Wolfgang Beyer,Kirsten Sandvig,Hiroyuki Yamamoto,Iris Lindberg,Lisa Walz,Veronika von Messling,Manuel E. Than,Wolfgang Garten,Torsten Steinmetzer +13 more
TL;DR: The results confirm that the inhibition of furin is a promising strategy for a short‐term treatment of acute infectious diseases.

Peptidomimetic furin inhibitor MI-701 in combination with oseltamivir and ribavirin efficiently blocks propagation of highly pathogenic avian influenza viruses and delays high level oseltamivir resistance in MDCK cells.
Yinghui Lu,Kornelia Hardes,Sven O. Dahms,Eva Böttcher-Friebertshäuser,Torsten Steinmetzer,Manuel E. Than,Hans-Dieter Klenk,Wolfgang Garten +7 more
TL;DR: It is indicated that combination with a potent furin inhibitor significantly enhances the therapeutic efficacy of conventional antivirals drugs against HPAIV infection.
32
New furin inhibitors based on weakly basic amidinohydrazones
TL;DR: A novel series of amidinohydrazone-derived furin inhibitors was prepared; the most potent compounds 17 and 21 inhibit furin with K(i) values of 0.46 and 0.59μM, respectively.
