Contaminated salad leaves have emerged as important vehicles for the transmission of enteric pathogens to humans. A recent outbreak of Salmonella enterica serovar Senftenberg (S. Senftenberg) in the United Kingdom has been traced to the consumption of contaminated basil. Using the outbreak strain of S. Senftenberg, we found that it binds to basil, lettuce, rocket and spinach leaves showing a pattern of diffuse adhesion. Flagella were seen linking S. Senftenberg to the leaf epidermis, and the deletion of fliC (encoding phase-1 flagella) resulted in a significantly reduced level of adhesion. In contrast, although flagella linking S. enterica serovar Typhimurium to the basil leaf epidermis were widespread, deletion of fliC did not affect leaf attachment levels. These results implicate the role of flagella in Salmonella leaf attachment and suggest that different Salmonella serovars use strain-specific mechanisms to attach to salad leaves.

Interaction of Salmonella enterica with basil and other salad leaves

The ability to subtype bacteria by typing methods provides the bacteriologist with a powerful means to identify relationships between bacteria. This knowledge is used to identify routes of disease transmission among livestock and from livestock to humans. In the present paper, the principles of bacterial typing and the most commonly applied typing methods for use in veterinary public health are discussed in the context of their application in the investigation of salmonella epidemiology. Typing methods are now routinely used in most investigations on this subject and have provided insight into routes of transmission, reservoirs of infection and mechanisms of persistent infection. Under the EC order on zoonotic diseases, extended surveillance on the presence of zoonotic bacteria in livestock must be expected. To receive the maximum benefits of this surveillance, selected typing methods must be applied to all isolates of e.g. salmonella. At present, serotyping, and phage typing where applicable, are the most obvious choices for continuous surveillance of this organism. Random amplification of polymorphic DNA (PCR based typing) may have the potential for allocating strains into relevant groups quicker and without the requirement for additional manpower, and this method may be preferred in future.

Bacterial typing methods suitable for epidemiological analysis. Applications in investigations of salmonellosis among livestock.

In September 1994, a complaint was registered at a public health unit concerning a cheese product. In addition, public health laboratories in Ontario reported an increase in the number of isolates of Salmonella berta from patients with diarrhoeal illness. A clinical, environmental and laboratory investigation was initiated to determine the nature of this outbreak. Isolates of Salmonella berta were compared using large fragment genomic fingerprinting by pulsed-field gel electrophoresis (PFGE). By late October, 82 clinical cases had been identified including 35 confirmed, 44 suspected and 3 secondary. The investigation linked illness to consumption of an unpasteurized soft cheese product produced on a farm and sold at farmers' markets. Subtyping results of patient, cheese and chicken isolates were indistinguishable, suggesting that the cheese was contaminated by chicken carcasses during production. The outbreak illustrates the potential role of uninspected home-based food producers and of cross-contamination in the transmission of foodborne bacterial pathogens.

A community outbreak of Salmonella berta associated with a soft cheese product.

This study explored the impact of feeding raw meat to dogs on the fecal prevalence of several enteric bacterial zoonotic pathogens. Campylobacter jejuni was isolated from 1/42 (2.6%) raw meat-fed dogs. Salmonella enterica was isolated from 2/40 (5%) of the raw meat feeds, 6/42 (14%) raw meat-fed dog feces, none of the dogs that did not receive raw meat (P = 0.001), 4/38 (10.5%) of the vacuum cleaner waste samples from households where raw meat was fed, and 2/44 (4.5%) of vacuum cleaner waste samples from households where raw meat was not fed to dogs (P = 0.41). Responses to a questionnaire probing practices and beliefs regarding raw meat feeding that was administered to dog owners demonstrated that dog owners may either not be aware or refuse to acknowledge the risks associated with raw meat-feeding; thus, they may neglect to conduct adequate intervention strategies to prevent zoonoses among themselves and their families.

Perceptions, practices, and consequences associated with foodborne pathogens and the feeding of raw meat to dogs.

Strains of Salmonella enterica serovar berta (S. berta) from Denmark and seven other countries have been characterized with the aim of developing a rational typing strategy in connection with outbreak investigations. Biotyping divided the strains into H2S-positive (90%) and H2S-negative (10%) biovars. Six percent of the strains were resistant to one or more antimicrobial agents. Eighty-eight percent of the strains carried plasmids and 52 different plasmid profiles were recognized. Six of the common plasmid sizes in these profiles were shown by restriction enzyme analyses to contain more than one plasmid species. More than 90% of the strains had the same ribotype with the restriction enzymes Sma I and EcoR I and the same whole cell protein profile. Outer membrane protein profiles and isoenzyme profiles were identical in all S. berta analysed. Plasmid profiling in combination with restriction enzyme analysis of plasmids seemed to be the most rational typing strategy for S. berta. The results indicated that S. berta strains regardless of geographical source or host are possibly clonal in nature.

Plasmid profiles of 674 Salmonella berta strains isolated in the period 1985-1989 from 674 cases of human infections were recorded. Five hundred and eighty-nine of the strains examined (87.4%) contained plasmids and the mean number of plasmids was 1.8 per strain. Sixteen different plasmid sizes were recognized, 13 of which had also been reported from S. berta of poultry origin. The plasmids formed 90 different profiles. Forty-five of these profiles, comprising 90.7% of isolates, had previously been recorded among poultry strains. Only 12 human isolates (1.8%) carried plasmids which were not demonstrated among isolates obtained from broilers. The prevalences of the 20 most commonly demonstrated profiles were not significantly different among isolates obtained from humans and broilers. The average difference between the prevalences of individual plasmid profiles was 2.1, with a standard deviation of 4.2. Based on the results, indications are that broilers represent the main reservoir for human S. berta infections in Denmark.

Salmonella enterica serovar berta was isolated for the first time in Denmark in October 1984. This serovar has subsequently become endemic in the broiler industry. Plasmid profile analysis of 610 strains of S. berta isolated from broilers and broiler parent stock in the period October 1984 to July 1989 showed that plasmids were present in 85.7% of strains investigated, with a mean of 1.8 plasmids per strain. A significant increase in the mean number of plasmids per strain was seen during the observation period. Seventeen different plasmid sizes and 90 different plasmid profiles were identified. Plasmid profiling of S. berta appears to represent a promising tool for analysing the epidemiology of S. berta infections in broilers.
