We report here complete 6-month results from the orbiting Space Environment Survivability of Living Organisms (SESLO) experiment. The world’s first and only long-duration live-biology cubesat experiment, SESLO was executed by one of two 10-cm cube-format payloads aboard the 5.5-kg O/OREOS (Organism/Organic Exposure to Orbital Stresses) free-flying nanosatellite, which launched to a 72°-inclination, 650-km Earth orbit in 2010. The SESLO experiment measured the long-term survival, germination, metabolic, and growth responses of Bacillus subtilis spores exposed to microgravity and ionizing radiation including heavy-ion bombardment. A pair of radiation dosimeters (RadFETs, i.e., radiation-sensitive field-effect transistors) within the SESLO payload provided an in-situ dose rate estimate of 6–7.6 mGy/day throughout the mission. Microwells containing samples of dried spores of a wild-type B. subtilis strain and a radiation-sensitive mutant deficient in Non-Homologoous End Joining (NHEJ) were rehydrated after 14, 91, and 181 days in space with nutrient medium containing with the redox dye alamarBlue (aB), which changes color upon reaction with cellular metabolites. Three-color transmitted light intensity measurements of all microwells were telemetered to Earth within days of each 24-hour growth experiment. At 14 and 91 days, spaceflight samples germinated, grew, and metabolized significantly more slowly than matching ground-control samples, as measured both by aB reduction and optical density changes; these rate differences notwithstanding, the final optical density attained was the same in both flight and ground samples. After 181 days in space, spore germination and growth appeared hindered and abnormal. We attribute the differences not to an effect of the space environment per se, as both spaceflight and ground-control samples exhibited the same behavior, but to a pair of ~15-day thermal excursions, after the 91-day measurement and before the 181-day experiment, that peaked above 46 °C in the SESLO payload. Because the payload hardware operated nominally at 181 days, the growth issues point to heat damage, most likely to component(s) of the growth medium (RPMI 1640 containing aB) or to biocompatibility issues caused by heat-accelerated outgassing or leaching of harmful compounds from components of the SESLO hardware and electronics.

Preliminary Development of a Fluidic Device for Astrobiology Missions

AbstractThe ability to detect and quantify organic molecules at extremely low concentrations makes the Surface-enhanced Raman Spectroscopy (SERS) an analytical technique increasingly employed in multiple fields of research, including biomedicine, agriculture, food, forensics and environmental monitoring. In recent years, the validation and operation of Raman spectrometers in planetary exploration missions paved the way toward the potential application of SERS beyond terrestrial domains. Aiming at compiling all relevant works published to date, the present review provides a detailed overview of the fields of space research in which the potential use of SERS is being investigated, including astrobiology and astronaut health monitoring. Surface-enhanced Raman spectroscopy (SERS) in planetary exploration and space research: a review of progress, challenges and opportunities

In light of an upcoming series of missions beyond low Earth orbit (LEO) through NASA’s Artemis program and the potential establishment of bases on the Moon and Mars, the effects of the deep space environment on biology need to be examined and protective countermeasures need to be developed. Even though many biological experiments have been performed in space since the 1960s, most of them have occurred in LEO and for only short periods of time. These LEO missions have studied many biological phenomena in a variety of model organisms, as well as utilized a broad range of technologies. Given the constraints of the deep space environment, however, future deep space biological missions will be limited to microbial organisms using miniaturized technologies. Small satellites like CubeSats are capable of querying relevant space environments using novel instruments and biosensors. CubeSats also provide a low-cost alternative to more complex and larger missions, and require minimal crew support, if any. Several have been deployed in LEO, but the next iteration of biological CubeSats will go farther. BioSentinel will be the first interplanetary CubeSat and the first biological study NASA has sent beyond Earth’s magnetosphere in 50 years. BioSentinel is an autonomous free-flyer platform able to support biology and to investigate the effects of radiation on a model organism in interplanetary deep space. The BioSensor payload contained within the free-flyer is also an adaptable instrument that can perform biologically relevant measurements with different microorganisms and in multiple space environments, including the ISS, lunar gateway, and on the surface of the Moon. Nanosatellites like BioSentinel can be used to study the effects of both reduced gravity and space radiation and can house different organisms or biosensors to answer specific scientific questions. Utilizing these biosensors will allow us to better understand the effects of the space environment on biology so humanity may return safely to deep space and venture farther than ever before.

Developing technologies for biological experiments in deep space

SporeSat – a lab-on-a-chip (LOC) centrifuge platform designed for integration as the payload of a small (5.5 kg), free-flying satellite – has been developed to determine the gravitational thresholds for calcium-ion channel activation of a single-cell spore from the fern Ceratopteris richardii. This fern is an important model system for gravity-directed plant-cell development during variable-gravity conditions attainable only in space flight. Calcium-ion channel activity is measured by photolithographically defined calcium ion–selective electrodes (ISEs) at opposite ends of each spore. Artificial gravity is created by rotating a disk-like platform that contains the spores in wells along with the calcium ISEs. Ground experiments reveal a maximum calcium concentration ratio at 2.2xg, between micro-ion-selective electrodes near the “top” and “bottom” ends of the spore, indicating an increasing calcium concentration at one “end” of the fern spore with respect to the other. Confocal micrographs of rhizoid formation confirm the light-induced germination. SporeSat is a spaceflight experiment that will take ~ 4 days; data will be telemetered to Earth over ~ 100 days.
