Purpose
It has been reported single-nucleotide polymorphisms (SNPs) of the IL-10 promoter might be associated with the susceptibility to recurrent pregnancy loss (RPL). Owing to the inconclusive results, we conducted a meta-analysis to systematically summarize and clarify the association between the IL-10 promoter SNPs and RPL risk.

Association of interleukin-10 gene promoter polymorphisms with recurrent pregnancy loss: a meta-analysis

Problem

This systematic review and meta-analysis aimed to synthesize and assess the correlation between the interleukin-10 (IL-10)-1082A/G polymorphism and idiopathic recurrent miscarriage (IRM) using relevant studies.



Method of study

Five electronic databases (Cochrane Library of Systematic Reviews, PubMed, EMBASE, Web of Science, and Google Scholar) were comprehensively searched up to August 2015. Data were extracted independently by two members of our team.



Results

The meta-analysis of 13 eligible studies suggested a recessive effect of the G allele (-1082A/G polymorphism) for IRM risk.



Conclusions

The pooled evidence demonstrated that the GG genotype increased IRM risk.

Cardiovascular disease (CVD) is the leading cause of  death in women in the Western world In this thesis, several studies are presented examining the association between recurrent miscarriage and cardiovascular disease Main aim of this thesis was to assess whether miscarriages are independently associated with an increased risk of cardiovascular disease later in life And, if this was true, to identify cardiovascular risk factors and predict long term cardiovascular disease risk in women with a history of recurrent miscarriage We found an increased risk of ischemic heart disease in women with a history of two (multivariate analysis HR 182) and three or more miscarriages (HR 318), irrespective whether consecutive or not (chapter 2) Women with a history of recurrent miscarriage have significantly higher 10- and 30-year cardiovascular risk scores compared to women with a history of no miscarriage These results indicate an opportunity for the early identification of women prone to cardiovascular disease later in life Women with a history of two or more miscarriages must be made aware of their increased cardiovascular risk and appropriate risk factor modifications will have to be offered, for example life style advises; weight management and smoking control

Impact of interleukin-10 promoter region polymorphisms on recurrent miscarriage: A case–Control approach

Karyotyping of 366 couples (732 individuals) with early recurrent pregnancy losses in anamnesis revealed chromosomal anomalies in 4.09% (30 cases)—within them 2.05% carry reciprocal translocations, in 0.82%-Robertsonian translocations, 0.55% carry numerical and structural gonosomal anomalies and in 0.27%—marker chromosome of unknown origin. The risk of early reproductive losses in women after excluding the cytogenetic component increases three fold if SNPs 1082GG, 592CC, 819CC of IL-10 gene and IFN-γ + 874AT or 874AA genotypes are present. ELISA-mediated detection of serum IL-10 and IFN-γ showed a possibly significant increase of IFN-γ in women with the history of early reproductive losses when compared to reproductively healthy women. We are proposing a complex cyto- and immunogenetic investigation in cases of early reproductive losses in women. One of the important issues of reproduction are the immunological mechanisms of pregnancy maintenance, where the disbalance in the genetically determined Th1- and Th2-cytokine levels may be one of the causes of early fetus elimination.
