The interaction specificity of salmon sperm DNA with various derivatives of daunorubicin has been studied. The results of binding, viscometric, 1H nuclear magnetic resonance (NMR), flow dichroism, DNA template inhibition, rates of dissociation, and circular dichroism studies are found to be consistent with an intercalation mode of binding of the anthracycline ring as has been shown by other investigators. Moreover, it is observed that (i) strength of binding, (ii) the ease of dissociation of DNA-anthracycline complexes, and (iii) the degree of inhibition of the DNA-dependent RNA polymerase are dependent on the presence of the amino sugar moiety of daunoseamine. The results are consistent with specific H bonding of the amino group of the sugar moiety with DNA as has been suggested earlier by Pigram et al. (Pigram, W.J., Fuller, W., and Hamilton, L.D. (1972), Nature (London), New Biol. 235, 17). Peptide derivatives substituted at the amino sugar function of daunorubicin lower the affinity of the drug to DNA and presumably interfere with the "full insertion" of the anthracycline drugs between base pairs of DNA. The significance of these findings in relation to the biological efficacy of daunorubicin and related derivatives as antileukemic agents is discussed.

Monitoring surgical wounds post-operatively is necessary to prevent infection, dehiscence and other complications. However, the monitoring of deep surgical sites is typically limited to indirect observations or to costly radiological investigations that often fail to detect complications before they become severe. Bioelectronic sensors could provide accurate and continuous monitoring from within the body, but the form factors of existing devices are not amenable to integration with sensitive wound tissues and to wireless data transmission. Here we show that multifilament surgical sutures functionalized with a conductive polymer and incorporating pledgets with capacitive sensors operated via radiofrequency identification can be used to monitor physicochemical states of deep surgical sites. We show in live pigs that the sutures can monitor wound integrity, gastric leakage and tissue micromotions, and in rodents that the healing outcomes are equivalent to those of medical-grade sutures. Battery-free wirelessly operated bioelectronic sutures may facilitate post-surgical monitoring in a wide range of interventions. Publisher Summary Daunomycin is an antitumor antibiotic of the anthracycline group, which was isolated in the Farmitalia Research Laboratories in Milan, Italy, from cultures of Streptomyces peucetius. The major part of the drug was found in the culture media of the mycelium and was isolated by means of solvent extraction, cation resins, and paper or column chromatography. The yield was 5–15 mg of product per liter of culture media. The soil sample was from Castel del Monte, near Puglie, and hence the name of the species S. peucetius. The drug is classified as an anthracycline and has similar physicochemical characteristics to rhodomycins, cynerubines, pyrromicins, and rutilantines. Daunomycin consists of a pigmented aglycone (daunomycinone) in glycoside linkage with an amino sugar (daunosamine). Daunomycin inhibits the multiplication of bacteriophage. It strongly inhibits DNA phages by acting during the replication cycle of the phage and not on absorption, injection, or lysis. The RNA phage growth was unaffected by daunomycin. The antiphage activity was proportional to the concentration of daunomycin. The antiphage activity was partially blocked by DNA but not by nucleotides or nucleosides.

The cardiac toxicity of daunomycin is probably related to the cumulative dose rather than to the length of time of treatment or the time interval since discontinuing therapy. This effect occurs more frequently and at lower doses than originally thought.A persistently increased cardiothoracic ratio may be a suggestive roentgenographic sign of a delayed manifestation of cardiac toxicity in patients receiving re-inducer courses of daunomycin for acute lymphocytic leukemia.This diagnosis should be considered in children receiving daunomycin who develop signs of cardiac failure after cessation of the drug.
