Imatinib mesylate (STI-571 Glivec, Gleevec) is an active agent for gastrointestinal stromal tumours, but does not yield responses in other soft-tissue sarcomas that are unselected for a molecular target. Results from an EORTC Soft Tissue and Bone Sarcoma Group phase II study.

TL;DR: In this article, the authors evaluated the antitumour response and time to progression (TTP) of patients treated with imatinib mesylate (Glivec®, Gleevec™, formerly STI-571) who had advanced and/or metastatic gastrointestinal stroma tumours (GIST) or other soft tissue sarcomas (STS).

TL;DR: It is concluded that a cross-over to high-dose imatinib is feasible and safe in GIST patients who progress on low-dose therapy.

ICI D1694, a Quinazoline Antifolate Thymidylate Synthase Inhibitor That Is a Potent Inhibitor of L1210 Tumor Cell Growth in Vitro and in Vivo: A New Agent for Clinical Study
Ann L. Jackman,G.A. Taylor,William Gibson,Kimbell R,M. Brown,Alan Hilary Calvert,Ian Judson,L. R. Hughes +7 more
TL;DR: The L1210:1565 cell line, which has greatly impaired reduced-folate/methotrexate transport and thus is resistant to methotrexate, was significantly cross-resistant to ICI D1694, suggesting that ICID1694 is dependent on this uptake mechanism for good cytotoxic potency in L 1210 cells.
