In the present study, surface plasmon resonance (SPR) based biosensor with SF-10 glass prism/zinc oxide/gold/MoS 2 /graphene hybrid structure is reported. The angular interrogation method is used for the reflectance spectra analysis. From the spectral analysis, we study the different performance parameters like sensitivity, detection accuracy, quality parameter and electric field intensity enhancement factor (EFIEF) of the proposed SPR based biosensor. Numerical results show that the base ZnO (zinc oxide) layer which have a large real value of the dielectric constant in combination with gold, MoS 2 and graphene, is responsible for enhancing the sensing performance of the proposed SPR based biosensor. The present biosensor can attain a maximum sensitivity (101.58 deg/RIU), detection accuracy (1.81) and quality parameter (15.11 RIU -1 ) for a large dynamic range of refractive index change from 1.33 to 1.45.

A study of surface plasmon resonance (SPR) based biosensor with improved sensitivity

We report that gold/zinc oxide (Au/ZnO) nanocomposite films were effectively employed to enhance the performance of surface plasmon resonance (SPR) for the detection of tumor markers. Carbohydrate antigen 15.3 (CA15-3), a tumor marker for breast cancer, was chosen as a model analyte. We analyzed intensity response to the samples at various concentrations (0.0125 U/mL to 160 U/mL) in pleural fluid to evaluate the detection capability of the SPR biosensor based on Au/ZnO thin films. The linear range extended from 1 to 40 U/mL with a correlation coefficient of R(2) = 0.991 and a limit of detection reaching 0.025 U/mL at a signal-to-noise ratio of 3:1. Compared with the degree of the shift in SPR intensity induced by the specific binding event between antibody and antigen, the change of intensity on the Au/ZnO layers was increased by at least 2 fold over that on the gold/chromium (Au/Cr) layers. In addition, we determined that the Au/ZnO layers allowed for a detection limit 4 times lower than the Au/Cr layers, which are in widespread use as the sensing interfaces in current SPR-based detectors. In conclusion, the use of Au/ZnO films greatly enhanced the SPR signal yield for this bimolecular interaction and showed high sensitivity.

High-sensitivity detection of carbohydrate antigen 15-3 using a gold/zinc oxide thin film surface plasmon resonance-based biosensor.

A nanoscale layer of chromium or titanium is commonly used in plasmonic nanoantennas to firmly adhere a goldfilm to a glass substrate, yet the influence of this layer on the antenna performance is often ignored. As a result, the need for the use of potentially better materials is not widely recognized. Using a single aperture milled in a goldfilm with 120 nm diameter as a nanobench for these investigations, we present thefirst experimental report of the strong dependence of the plasmonic enhancement of single-moleculefluorescence on the nature of the adhesion layer. By combiningfluorescence correlation spectroscopy andfluorescence lifetime measurements,weshowthatthisstructureisverysensitivetothepropertiesoftheadhesionlayer,andwedetail therespectivecontributionsofexcitationandemissiongainstotheobservedenhancedfluorescence.Anyincrease in the absorption losses due to the adhesion layer permittivity or thickness is shown to lower the gains in both excitation and emission, which we relate to a damping of the energy coupling at the nanoaperture. With this nanobench, we demonstrate the largest enhancement factor reported to date (25) by using a TiO2adhesion layer. The experimental data are supported by numerical simulations and argue for a careful consideration of the adhesion layer while designing nanoantennas for high-efficiency single-molecule analysis.

Crucial Role of the Adhesion Layer on the Plasmonic Fluorescence

Thin-film gold sensor substrates with 2 nm chromium and titanium adhesion layers have been characterised in the thickness range of 30–70 nm. We correlate the intensity of the optical transmission peak near 500 nm with the amplitude of the surface plasmon resonance (SPR) response in water. The optical transmission is demonstrated to be a better control for manufacturing chips than (quartz crystal monitoring) QCM and transmission and absorbance ranges for acceptable SPR response are given. We demonstrate the use of disposable gold chips manufactured from readily available No. 3 cover-glass substrates which can be used on both portable SPR instruments and on commercial machines such as Biacore. Ti/Au substrates are preferred over Cr/Au based on the lower optical absorbance of titanium adhesion layers.

Characterisation of gold surface plasmon resonance sensor substrates

In the present communication, a highly sensitive surface plasmon resonance (SPR) biosensor with Kretschmann configuration having alternate layers, prism/zinc oxide/silver/gold/graphene/biomolecules (ss-DNA) is presented. The optimization of the proposed configuration has been accomplished by keeping the constant thickness of zinc oxide (32 nm), silver (32 nm), graphene (0.34 nm) layer and biomolecules (100 nm) for different values of gold layer thickness (1, 3 and 5 nm). The sensitivity of the proposed SPR biosensor has been demonstrated for a number of design parameters such as gold layer thickness, number of graphene layer, refractive index of biomolecules and the thickness of biomolecules layer. SPR biosensor with optimized geometry has greater sensitivity (66 deg/RIU) than the conventional (52 deg/RIU) as well as other graphene-based (53.2 deg/RIU) SPR biosensor. The effect of zinc oxide layer thickness on the sensitivity of SPR biosensor has also been analysed. From the analysis, it is found that the sensitivity increases significantly by increasing the thickness of zinc oxide layer. It means zinc oxide intermediate layer plays an important role to improve the sensitivity of the biosensor. The sensitivity of SPR biosensor also increases by increasing the number of graphene layer (upto nine layer).
