Given the problems of species delimitation in algae using morphology or sexual compatibility, molecular data are becoming the standard for delimiting species and testing their traditional boundaries. The idea that species are separately evolving metapopulation lineages, along with theoretical progress in phylogenetic and population genetic analyses, has led to the development of new methods of species delimitation. We review these recent developments in DNA-based species delimitation methods, and discuss how they have changed and continue to change our understanding of algal species boundaries. Taxonomy of the brown algal genus Dictyota has a long and troubled history. Our inability to distinguish morphological plasticity from fixed diagnostic traits that separate the various species has severely confounded species delineation. From continental Europe, more than 60 species and intraspecific taxa have been described over the last two centuries. Using a molecular approach, we addressed the diversity of the genus in European waters and made necessary taxonomic changes. A densely sampled DNA data set demonstrated the presence of six evolutionarily significant units (ESUs): Dictyota dichotoma (Huds.) J. V. Lamour., D. fasciola (Roth) J. V. Lamour., D. implexa J. V. Lamour., D. mediterranea (Schiffn.) G. Furnari, D. spiralis Mont., and the newly described D. cyanoloma sp. nov., which was previously reported as D. ciliolata from the Mediterranean Sea. Species distributions, based on DNA-confirmed occurrence records, indicate that all species are geographically confined to the NE Atlantic Ocean with the exception of D. dichotoma and D. implexa, which also occur in South Africa and Bermuda, respectively. To investigate potential hybridization between D. dichotoma and D. implexa, which were previously shown to be sexually compatible in culture, we compiled and analyzed sets of mitochondrial, plastid, and nuclear markers to detect putative hybrids or introgression in natural populations. Failure to detect natural hybrids indicates that effective pre- and postzygotic isolation mechanisms are at play in natural populations and supports the by-product hypothesis of reproductive isolation.

Species delimitation, taxonomy, and biogeography of dictyota in europe (dictyotales, phaeophyceae)

During secondary contact between phylogenetically closely related species (sibling species) having diverged in allopatry, the maintenance of species integrity depends on intrinsic and extrinsic reproductive barriers. In kelps (Phaeophyceae), the observations of hybrids in laboratory conditions suggest that reproductive isolation is incomplete. However, not all interspecific crosses are successful, and very few hybrids have been observed in nature, despite the co-occurrence of many kelp species in sympatry. This suggests that there are reproductive barriers that maintain species integrity. In this study, we characterized the fine genetic structure of a secondary contact zone to clarify the extent of reproductive isolation between two sister species. In Lessonia nigrescens Bory (Laminariales, Phaeophyta) species complex, two cryptic species have been recently found out from gene phylogenies, and-waiting for a formal taxonomic description-we used their geographic distribution to name them (northern and southern species). We studied 12 populations, distributed along 50 km of coastline, and employed two molecular approaches, assigning individuals to phylogenetic species according to a diagnostic mitochondrial marker (351 individuals analyzed) and quantifying interspecific gene flow with four microsatellite markers (248 individuals analyzed). No hybridization or introgression was revealed, indicating complete reproductive isolation in natural conditions. Unexpectedly, our study demonstrated that the two species were strictly segregated in space. This absence of co-occurrence along the contact zone can partially explain the lack of hybridization, raising new interesting questions as to the mechanisms that limit sympatry at small spatial scales.

Molecular markers belonging to the three different genomes, mitochondrial (cox2-cox3 spacer), plastid (rbcL), and nuclear (internal transcribed spacer [ITS] 2 region), were used to compare samples of the two morphologically related species Gracilaria gracilis (Stackh.) Steentoft, L. M. Irvine et Farnham and G. dura (C. Agardh) J. Agardh collected along Atlantic coasts. In northern Europe, the distinction between these two species is ambiguous, and they are currently recognized under the single name of G. gracilis. The low but congruent patterns of genetic divergence observed for markers of the three genomic compartments highly suggest that these two taxa correspond effectively to two different genetic entities as previously described 200 years ago, based on morphological traits. However, thanks to the combination of different DNA markers, occurrence of “incongruent” cytotypes (i.e., mitotypes of G. dura associated with chlorotypes of G. gracilis) in individuals collected from Brittany, suggests interspecific hybridization between the two sibling species studied.

SUMMARY

Phylogeographic signal provided by the newly developed 23S plastid rRNA marker (universal plastid amplicon, UPA) and the cytochrome oxidase subunit I gene marker (COI) in the freshwater red alga Batrachospermum helminthosum, throughout its range in North America, was investigated. These markers were compared in individuals from a previous study using the cytochrome oxidase 2–3 spacer region (cox2–3), which has yielded the most useful data to date with 13 haplotypes among geographic locations. Five haplotypes were resolved for the UPA, differing by only one to two base pairs (bp), and we conclude that this marker may be more appropriate for studying interspecific variation. In contrast, the COI gene revealed 16 haplotypes, differing from one to 44 base pairs or up to 6.6% sequence variation. The intraspecific variation of COI in this taxon is much greater than that reported thus far for marine red algae (generally <5 bp). The intraspecific variation within B. helminthosum is in accord with levels shown in Batrachospermum macrosporum (48 bp within distant locations in Brazil). The COI gene is comparable to the cox2–3 spacer for phylogeographic studies as the haplotype networks were similar and showed the same geographic patterns. To our knowledge, this is the first comparison of these three regions for phylogeographic research in the red algae.

Mastocarpus stellatus is a common intertidal red alga in the north Atlantic. Previous work showed that the species was divided into at least two breeding groups in the northeastern Atlantic, a northern one (absent in Spain and Portugal) and a southern one, which were almost completely inter-sterile. It also showed that, in laboratory culture, at least two types of life history were evident in this species (a sexual one and a ‘direct’ one). The distribution of these life histories appeared to have a north–south distribution. We have used molecular markers to confirm these results and expand the sampling in certain populations. Organellar markers showed that the breeding groups have different plastid and mitochondria haplotypes and appear to be distributed along a north–south gradient. Populations in southern England and northern France (Brittany) have mixed northern and southern breeding groups, except for the Rade du Brest, as was shown previously from culture studies.
