Parasitic diseases of sheep involving gastrointestinal nematodes (GIN) are one of the main problems that affect fl ock productivity, especially during the peripartum period. Around lambing, the ewes are immunosuppressed and the nematode faecal egg count (FEC) increases at four weeks before lambing, reaching a peak between the fourth and sixth week postpartum and subsequently decreasing towards weaning. Prolactin has been credited with a suppressive effect on immune system, along with other hormones that intervene in metabolism, such as leptin, which has an important role in the activation of other hormones. Cortisol has also been included; this is stimulated by any stressful event and inhibits the proliferation of T-cells and alters the function of immunoglobulins. Another related hormone is pepsinogen, which is considered a marker of the integrity of the abomasum mucosa, as well as the albumin concentration that increases in the presence of a GIN infection. The humoral and cellular immune response, as well as inflammatory reactions, are the main mechanisms of action against GIN. Lymphocytes direct the effector mechanisms in a Th2 cell response, including interleukins (IL-4, IL-5, IL-9, IL-10) and transforming growth factor beta (TGF-β) together with immunoglobulins (IgA, IgG, IgM and IgE), which prevent the invasion of pathogens. Eosinophils with a cytotoxic effect are indicators of a parasitic infection, with importance in the immune protection of infected individuals. The genetic selection of resistant individuals measured by FEC is dependent on the heritability (h2), which is moderately inheritable and highly repeatable. Effects that influence the resistance or susceptibility of sheep to GIN infections in the peripartum period are determined by the interaction of various factors, such as genotype or breed, nutrition, age, type of birth, season and production system, etc., which are studied in this review.

This study estimated genetic parameters and identified candidate genes associated with live weight, and the occurrence of pregnancy in 1327 Romney ewe lambs using genome-wide association studies. Phenotypic traits considered were the occurrence of pregnancy in ewe lambs and live weight at eight months of age. Genetic parameters were estimated, and genomic variation was assessed using 13,500 single-nucleotide polymorphic markers (SNPs). Ewe lamb live weight had medium genomic heritability and was positively genetically correlated with occurrence of pregnancy. This suggests that selection for heavier ewe lambs is possible and would likely improve the occurrence of pregnancy in ewe lambs. No SNPs were associated with the occurrence of pregnancy; however, three candidate genes were associated with ewe lamb live weight. Tenascin C (TNC), TNF superfamily member 8 (TNFSF8) and Collagen type XXVIII alpha 1 chain (COL28A1) are involved in extracellular matrix organization and regulation of cell fate in the immune system. TNC may be involved in ewe lamb growth, and therefore, could be of interest for selection of ewe lamb replacements. The association between ewe lamb live weight and TNFSF8 and COL28A1 is unclear. Further research is needed using a larger population to determine whether the genes identified can be used for genomic selection of replacement ewe lambs.

Genome-Wide Association Studies of Live Weight at First Breeding at Eight Months of Age and Pregnancy Status of Ewe Lambs

Parasite infections can have substantial impacts on population dynamics and are accordingly a key challenge for wild population management. Here we studied genetic mechanisms driving parasite resistance in a large herbivore through a comprehensive approach combining measurements of neutral (16 microsatellites) and adaptive (MHC DRB1 exon 2) genetic diversity and two types of gastrointestinal parasites (nematodes and coccidia). While accounting for other extrinsic and intrinsic predictors known to impact parasite load, we show that both neutral genetic diversity and DRB1 are associated with resistance to gastrointestinal nematodes. Intermediate levels of multi-locus heterozygosity maximized nematodes resistance, suggesting that both in- and outbreeding depression might occur in the population. DRB1 heterozygosity and specific alleles effects were detected, suggesting the occurrence of heterozygote advantage, rare-allele effects and/or fluctuating selection. On the contrary, no association was detected between genetic diversity and resistance to coccidia, indicating that different parasite classes are impacted by different genetic drivers. This study provides important insights for large herbivores and wild sheep pathogen management, and in particular suggests that factors likely to impact genetic diversity and allelic frequencies, including global changes, are also expected to impact parasite resistance.

Association between variation in faecal egg count for a natural mixed field-challenge of nematode parasites and TLR4 variation.

Flystrike is a major cost and a welfare issue for the New Zealand sheep industry. There are several factors that can predispose sheep to flystrike, such as having fleecerot, a urine-stained breech, and ‘dags’ (an accumulation of faecal matter in the wool of the breech). The FABP4 gene (FABP4) has been associated with variation in ovine fleecerot resistance, with a strong genetic correlation existing between fleecerot and flystrike occurrence. In this study, blood samples were collected from sheep with and without flystrike for DNA typing. PCR-SSCP analyses were used to genotype two regions of ovine FABP4. Sheep with the A1 variant of FABP4 were found to be less likely (odds ratio 0.689, P = 0.014) to have flystrike than those without A1. The likelihood of flystrike occurrence decreased as copy number of A1 increased (odds ratio 0.695, P = 0.006). This suggests that FABP4 might be a candidate gene for flystrike resilience in sheep, although further research is required to verify this association.
