Natural regulatory mutations elevate the fetal globin gene via disruption of BCL11A or ZBTB7A binding
Gabriella E. Martyn,Beeke Wienert,Lu Yang,Manan Shah,Laura J. Norton,Jon Burdach,Ryo Kurita,Yukio Nakamura,Richard C. M. Pearson,Alister P. W. Funnell,Kate G. R. Quinlan,Merlin Crossley +11 more
TL;DR: It is found that the major fetal globin gene repressors BCL11A and ZBTB7A directly bound to the sites at –115 and –200 bp, respectively, and introduced of naturally occurring HPFH-associated mutations into erythroid cells by CRISPR–Cas9 disrupted repressor binding and raised γ-globin gene expression.

The miR-100-mediated pathway regulates apoptosis against virus infection in shrimp.
Lu Yang,Geng Yang,Xiaobo Zhang +2 more
TL;DR: The results indicated that the silencing of miR-100 expression resulted in the increase of apoptotic activity of shrimp hemocytes and further led to the decreases of virus genome copies in shrimp and virus-infected shrimp mortality compared with the controls.

Methylation of a CGATA element inhibits binding and regulation by GATA-1.
Lu Yang,Zhiliang Chen,Elizabeth S. Stout,Fabien Delerue,Fabien Delerue,Lars M. Ittner,Lars M. Ittner,Marc R. Wilkins,Kate G. R. Quinlan,Merlin Crossley +9 more
TL;DR: The authors show that methylation of a single CGATA element within the c-Kit gene inhibits binding and regulation by erythroid transcription factor GATA-1, both in cells and in mice, suggesting that methylations at this site plays an essential role in erythropoiesis.

Specific base modification antisense nucleotide chain and application thereof
Geng Yang,Jiajia Wang,Lu Yang,Xiaobo Zhang +3 more
- 21 Nov 2012
TL;DR: In this paper, a short chain DNA of 16 nt has been selected through specific base modification, which enables degradation of the short-chain DNA byin vivo DNA enzyme to be effectively prevented and action time of short-chains DNA to be prolonged, which can specifically knock down the expression of miRNA-100 without affecting the expression level of other miRNAs.

Application of miR-100 inhibitor in preparation of medicine for treating breast cancer
Zhang Xiaobo,Gong Yi,Lu Yang +2 more
- 02 Jul 2014
TL;DR: The miR-100 inhibitor disclosed by the invention can induce apoptosis of tumor cells, growth of tumors in tumor transplanted mice can be effectively induced, the sensitivity of breast cancer cells on chemotherapy medicines is increased, the effect of chemotherapy is improved, and the miR100 is a latent new target for the medicine for treating the breast cancer, and is a new path for medical therapy.
