Gating TrkB switch by methylglyoxal enables GLO1 as a target for depression
Ziyin Wu,Ziyin Wu,Yingxue Fu,Yinfeng Yang,Chao Huang,Chunli Zheng,Zihu Guo,Zihao Yang,Xuetong Chen,Jinglin Zhu,Jinghui Wang,Xiaogang Li,Liyang Chen,Weiwei Zhou,Yangyang Chen,J.G. Wang,Yang Yang,Meng Jiang,Su-Shing Chen,Aiping Lu,Jianlin Liu,Yan Li,Shiguo Sun,Zhenzhong Wang,Wei Xiao,Yonghua Wang +25 more
TL;DR: A novel therapeutic axis of methylglyoxal (MGO)/tropomyosin receptor kinase B (TrkB) for treating major depressive disorder is reported and principles for the rational design of new antidepressants targeting GLO1 are depicted.

Methylglyoxal modification of TrkB promotes synaptic plasticity and enhances resilience to chronic stress
Ziyin Wu,Ziyin Wu,Yingxue Fu,Yinfeng Yang,Chao Huang,Chunli Zheng,Zihu Guo,Zihao Yang,Xuetong Chen,Jinglin Zhu,Jinghui Wang,Xiaogang Li,Liyang Chen,Weiwei Zhou,Yangyang Chen,J.G. Wang,Yang Yang,Meng Jiang,Su-Shing Chen,Aiping Lu,Jianlin Liu,Yan Li,Shiguo Sun,Zhenzhong Wang,Wei Xiao,Yonghua Wang +25 more
TL;DR: It is found that MGO concentrations are decreased in the prefrontal cortex and hippocampus in rats subjected to chronic stress, and low-dose MGO treatment remarkedly enhances resilience to stress and alleviates depression-like symptoms.
Systems-level analysis identifies key regulators driving epileptogenesis in temporal lobe epilepsy
Yingxue Fu,Zihu Guo,Ziyin Wu,Liyang Chen,Yaohua Ma,Zhenzhong Wang,Wei Xiao,Yonghua Wang,Yonghua Wang +8 more
TL;DR: A systems-level analytic framework is built which integrates gene meta-signatures, gene coexpression network and cellular regulatory network to unveil the evolution landscape of epileptogenic process and to identify key regulators that govern the transition between different epileptogenesis stages.

Systems pharmacology uncovers serotonergic pathway mediated psychotherapeutic effects of Lonicerae Japonicae Flos
Ziyin Wu,Liyang Chen,Zihu Guo,Kaiyue Li,Yingxue Fu,Jinglin Zhu,Xuetong Chen,Chao Huang,Chunli Zheng,Yaohua Ma,Xiaogang Li,Jun Zhou,Zhenzhong Wang,Wei Xiao,Yonghua Wang +14 more
TL;DR: LJF and its active ingredient luteolin can reversed depressive/anxiety-like behavior in rats exposed to chronic mild stress (CMS), restored abnormal gene expression changes and neurotransmission in the striatum of CMS rats, and these effects were mediated through triggering the activation of the HTR2A/PLCγ/ERK/CREB pathway.

Systems-level analysis identifies key regulators driving epileptogenesis in temporal lobe epilepsy.
TL;DR: The upregulation of FAM107A, LAMB2, LTBP1 and TGIF1, which are mainly involved in nervous system development, were found contributing to both conditions and provide candidate regulators that may serve as potential antiepileptogenic targets.
