The present invention discloses a 3D printing bio-ink based on an acellular costal cartilage matrix and a preparation method and an application of the 3D printing bio-ink, and belongs to the technicalfield of biomedical materials. The invention discloses a tissue engineering meniscus scaffold and a preparation method thereof The method comprises the following steps: constructing a three-dimensional data model of the initial shape of a meniscus to be repaired; providing bio-ink, wherein the bio-ink comprises a hydrogel material, a matrix material, a curing agent and seed cells; providing a degradable polymer material; and with biological ink as a raw material of hydrogel fibers, a degradable high polymer material as a raw material of degradable high polymer material fibers, printing the scaffold according to the three-dimensional data model, wherein the scaffold is C-shaped and is consistent with the initial shape of the meniscus to be repaired, the scaffold comprises a plurality of scaffold layers sequentially stacked in the height direction of the scaffold, each scaffold layer comprises a plurality of degradable high polymer material fibers arranged at intervals and hydrogel fibers located in gaps among the degradable high polymer material fibers, and the degradable high polymer material fibers of every two adjacent scaffold layers intersect with each other
