Salicylic acid based small molecule inhibitor for the oncogenic Src homology-2 domain containing protein tyrosine phosphatase-2 (SHP2).
Xian Zhang,Yantao He,Sijiu Liu,Zhi-Hong Yu,Zhong Xing Jiang,Zhenyun Yang,Yuanshu Dong,Sarah C. Nabinger,Li Wu,Andrea M. Gunawan,Lina Wang,Rebecca J. Chan,Zhong Yin Zhang +12 more
TL;DR: A salicylic acid based combinatorial library approach aimed at binding both active site and unique nearby subpockets for enhanced affinity and selectivity led to the identification of a SHP2 inhibitor II-B08 (compound 9) with highly efficacious cellular activity, providing supporting evidence that chemical inhibition ofSHP2 may be therapeutically useful for anticancer and antileukemia treatment.

Acquisition of a Specific and Potent PTP1B Inhibitor from a Novel Combinatorial Library and Screening Procedure
TL;DR: A combinatorial approach that is designed to target both the active site and a unique peripheral site in PTP1B, demonstrating that it is possible to acquire potent, yet highly selective inhibitors for individual members of the large PTPase family of enzymes.

The Cys(X)5Arg Catalytic Motif in Phosphoester Hydrolysis
Zhong Yin Zhang,Yuan Wang,Li Wu,Li Wu,Eric B. Fauman,Jeanne A. Stuckey,Heidi L. Schubert,Mark A. Saper,Jack E. Dixon +8 more
TL;DR: It is shown that Arg409 plays a critical role in PTPase action and that the Cys(X)5Arg active site motif forms a phosphate-binding loop which appears to represent the essential features necessary for catalysis by the PTPases, the dual specific phosphatase, and the low molecular weight acid phosphatases.

TL;DR: Using a structure-guided and fragment-based library approach, a novel hydroxyindole carboxylic acid-based SHP2 inhibitor 11a-1 is identified, with an IC50 value of 200 nM and greater than 5-fold selectivity against 20 mammalian PTPs.

Structural basis of plasticity in protein tyrosine phosphatase 1B substrate recognition.
Mauro Sarmiento,Yoram A. Puius,Stefan W. Vetter,Yen Fang Keng,Li Wu,Yu Zhao,David S. Lawrence,Steven C. Almo,Zhong Yin Zhang +8 more
TL;DR: The crystal structures of PTP1B in complex with Ac-ELEFpYMDYE-NH(2) (PTP1 B.Con) and Ac-DAD(Bpa)pYLIPQQG (P TP1B.B Pa) are reported to show how aromatic/aliphatic residues at the -1 and -3 positions of peptide substrates are accommodated by PTP 1B.
