Abstract P4-03-16: Cyclin E affects Smad3 pathway in trastuzumab resistant HER2+ breast cancer
TL;DR: Inhibition of cyclin E activity with CDK2 inhibitor therapy (CDK2i, 600 nM) significantly decreased BT474R2 cell proliferation and results in trastuzumab resistance in HER2+ breast cancer.

Synergistic effect of eribulin and CDK inhibition for the treatment of triple negative breast cancer
Shreyas S. Rao,Jenna R. Stoehr,Danijela Dokic,Lei Wan,Joseph T. Decker,Kristine E. Konopka,Alexandra L. Thomas,Jia Wu,Virginia G. Kaklamani,Lonnie D. Shea,Lonnie D. Shea,Jacqueline S. Jeruss +11 more
TL;DR: The potential of this novel drug combination, CYC065 and eribulin, to suppress the growth of TNBC cells in vitro and in vivo is highlighted, warranting further clinical investigation.
Cyclin E overexpression confers resistance to trastuzumab through noncanonical phosphorylation of SMAD3 in HER2+ breast cancer.
Joseph T. Decker,Pridvi Kandagatla,Pridvi Kandagatla,Lei Wan,Regan Bernstein,Jeffrey A. Ma,Lonnie D. Shea,Jacqueline S. Jeruss +7 more
TL;DR: For patients with HER2+ breast cancer, a mechanism of CCNE-mediated trastuzumab resistance, regulated through noncanonical SMAD3 phosphorylation, could be treated with CDK2 inhibition to help enhance the efficacy of trastzumab therapy.
