Dale W. Garsed,Ahwan Pandey,Sian Fereday,Catherine Kennedy,Kazuaki Takahashi,Kathryn Alsop,Phineas T. Hamilton,Joy Hendley,Yoke-Eng Chiew,Nadia Traficante,Pamela J. Provan,Dinuka Ariyaratne,George Au-Yeung,D. Nicholas Bateman,Leanne Bowes,Alison Brand,Elizabeth L. Christie,Julie M. Cunningham,Michael Friedlander,Bronwyn Grout,Paul R. Harnett,Jillian Hung,Bryan M. McCauley,Orla McNally,Anna M. Piskorz,Flurina Saner,Robert A. Vierkant,Chen Wang,Stacey J. Winham,Paul D.P. Pharoah,James D. Brenton,Thomas P. Conrads,G. Larry Maxwell,Susan J. Ramus,Celeste Leigh Pearce,Malcolm C. Pike,Brad H. Nelson,Ellen L. Goode,Antoinette DeFazio,David D.L. Bowtell +39 more
TL;DR: Tumors of long-term survivors were more likely to have multiple alterations in genes associated with DNA repair and more frequent somatic variants resulting in an increased predicted neoantigen load, and patients clustered into survival groups based on genomic and immune cell signatures, including three subsets of patients with BRCA1 alterations with distinctly different outcomes.

TL;DR: It is demonstrated that IFNε is an intrinsic tumour suppressor in the female reproductive tract whose activities in models of established and advanced ovarian cancer, distinct from other type I IFNs, are compelling indications of potential new therapeutic approaches for ovarian cancer.

Harnett,Viola Heinzelmann-Schwarz,Michael Friedlander,Orla McNally,Michael A. Quinn,Peter J. Campbell,Anna deFazio,David D.L. Bowtell,David D.L. Bowtell +29 more
TL;DR: The findings underscore the large degree of variation in DNA copy number in spatially and temporally separated tumors in HGSC patients, and define LRP1B as a potential contributor to the emergence of chemotherapy resistance in these patients.
Small-scale mutations are infrequent as mechanisms of resistance in post-PARP inhibitor tumour samples in high grade serous ovarian cancer

TL;DR: It is indicated that mutations in genes related to DNA repair are detected in a minority of HGSC patients as genetic mechanisms of resistance, and future research into resistance in HGSC should focus on copy number, transcriptional and epigenetic aberrations, and the contribution of the tumour microenvironment.
EP362/#696 New patient-derived models and therapy screening in mucinous ovarian carcinoma
Olivia Craig,Carolina Salazar,Suad M. Abdirahman,Niveditha Rajadevan,Xiang Li,Kaylene Simpson,Richard Lupat,Leanne Bowes,Nadia Traficante,Patricia Wojtowicz,Jessica Boros,Catherine Kennedy,Anna De Fazio,Orla Mcnally,Clare Scott,Kylie Gorringe +15 more
- 01 Nov 2023
TL;DR: Researchers developed patient-derived 3D organoid models of mucinous ovarian carcinoma and screened 14 therapies, finding limited response to platinum-based chemotherapies, but promising results with topoisomerase I inhibitors and other agents, suggesting alternative treatment options.
