Bone diseases and fractures are uprising health burden in modern era Natural bone healing process is time consuming process, therefore bone grafting techniques were introduced to recover quickly Autograft was considered as the Gold Standard for past decades, but lack of donor sites enforced for alternative methods such as allograft and xenograft These methods were also restricted by limited range of applications and ethical constraints Bone substitutes were able fill up the voids in bone grafting methods and became more effective Poor mechanical properties and uncontrollable degradation made restrictions on bone treatments Bone Tissue Engineering have become the emerging method in nowadays which showed promising signs of healing bone diseases and defects including tumours However, there are challenges to overcome, this process will discover a cure in future for the betterment of the mankind

A Review on Bone Grafting, Bone Substitutes and Bone Tissue Engineering

Introduction Many clinical situations, such as spinal arthodesis, total joint arthroplasty, osteoprotic insufficiency fractures, and bone loss after skeletal trauma might be amenable to treatment options which include biodegradable scaffolds that can be injected and crosslinked in-situ to fill a defect. For minimally invasive applications, injectable systems that can be crosslinked in-situ by chemical redox initiation and can promote migration and proliferation of osteoblasts from the surrounding tissues would be attractive [1]. We recently developed a poly(caprolactonefumarate) (PCLF) macromer that self-crosslinks in the absence of a crosslinking agent. Above its melting point of 45-55C, the copolymer is a viscous liquid and it can be used as an injectable matrix to fill irregularly shaped defects [2]. At physiological temperature, the macromer self-crosslinks and hardens in-situ physically by crystallization and chemically by radical polymerization. The objective of this work was to study migration and proliferation of osteoblasts in porous PCLF scaffolds in vitro.
