Dynamics of CRISPR-Cas9 genome interrogation in living cells
Spencer C. Knight,Liangqi Xie,Wulan Deng,Benjamin Guglielmi,Lea B. Witkowsky,Lana Bosanac,Elisa T. Zhang,Mohamed El Beheiry,Jean-Baptiste Masson,Maxime Dahan,Maxime Dahan,Zhe Liu,Jennifer A. Doudna,Robert Tjian +13 more
TL;DR: It is shown that three-dimensional diffusion dominates Cas9 searching in vivo, and off-target binding events are, on average, short-lived (<1 second).

Efficient optical trapping and visualization of silver nanoparticles.
TL;DR: Efficient optical trapping combined with sensitive optical detection of individual silver nanoparticles was performed and it was found that the larger the particle, the stronger the optical force.

Ruda,Timothée Lionnet,Timothée Lionnet,Daniel Zenklusen,Daniel Zenklusen,Benjamin Guglielmi,Benjamin Guglielmi,Robert Tjian,Robert Tjian,Robert H. Singer +21 more
TL;DR: The current state of the field and the progress necessary to achieve a more complete understanding of the various steps in transcription are reported on.
Single-molecule tracking in live cells reveals distinct target-search strategies of transcription factors
Xavier Darzacq Dugast-Darzacq,Florence Proux,Olivier Bénichou,Raphaël Voituriez,Olivier Bensaude,Lana Bosanac,Lydia Boudarene +6 more
- 01 Jan 2014
TL;DR: The geometry-controlled kinetics of TFs target-search illustrates the influence of nuclear architecture on gene regulation, and has strong implications on how proteins react in the nucleus and how their function can be regulated in space and time.

Stretching short DNA tethers using optical tweezers
TL;DR: In this article, the authors used optical tweezers to measure the force-extension curves of double-stranded DNA and showed that these curves are similar to those predicted by WLC theories, but a reasonable fit can only be made if the persistence length is allowed to be a fitting parameter.
