Minimizing genetic adaptation in captive breeding programs: A review

Characterization of urinary and fecal metabolites of testosterone and their measurement for assessing gonadal endocrine function in male nonhuman primates.

This essay explains the role of reproductive science, including what are termed reproductive technologies (i.e. artificial insemination, in vitro fertilization, embryo transfer, cloning), in conservation biology. Reproductive techniques (high- and low-tech) find their greatest application in understanding species uniqueness, adaptations and physiological mechanisms, not in the large-scale assisted breeding and the production of offspring. Models of how to use these tools to study reproductive fitness are emerging to help insure gene diversity and even propagate endangered species, but only after fundamental databases have been developed. Examples are provided of how non-invasive hormone metabolite monitoring, artificial insemination and genome resource banking are being used ex situ and in situ to understand wildlife biology. We predict that as the fundamental, multi-species database grows, so will the applied benefits for: (1) developing genome banks for insuring extant genetic diversity; (2) assessing the relationship of physiology, behaviour and environmental perturbations; (3) managing small populations; and (4) dealing with dilemmas ranging from contraception to skewed sex ratios to animal welfare. Most progress will be made in using these tools in systematic studies to solve the mystery of how thousands of unstudied species reproduce. Carried out appropriately, financial costs will be consistent with any approach for generating sound scientific knowledge.

Sex and wildlife: the role of reproductive science in conservation

Most conventional spermatology research involves common mammalian species including livestock, laboratory animals and humans. Yet, there are more than 4500 mammalian species inhabiting the planet for which little is known about basic reproductive biology, including sperm characteristics and function. This information is important, not just as adjunct knowledge, but because the majority of these species are threatened with extinction, largely due to human-induced pressures. The field of conservation is changing rapidly, and global cooperation is emerging among a variety of wildlife enthusiasts, ranging from management authorities of nature reserves to curators of rare zoological collections. Conservation progress depends on systematic, multidisciplinary research first to answer basic questions, with new data then applied to endangered species management plans. The reproductive physiologist is a crucial component of this scheme. Reproduction is the essence of species survival, and enormous effort needs to be directed at these 'untraditional' research species, subspecies and populations. Spermatology research combined with simultaneous efforts in endocrinology, embryology and cryopreservation (among others) can lead to the successful application of assisted reproduction. Examples from this laboratory include an array of wild felid species and a rare cervid and mustelid. Obstacles to success are formidable, including unique species-specificities, diminished genetic diversity and a general lack of resources. Nonetheless, the field offers tremendous opportunities for generating unique knowledge of comparative interest and with conservation utility.

Spermatology for Understanding, Managing and Conserving Rare Species

To determine the primary excretory by-products of testosterone (T), 85 microCi [3H]T was administered i.v. to two adult Eld's deer stags. Blood (10 ml) was collected by jugular venipuncture at 0, 5, 10, 15, 30, 45, 60, 90, 120, 150, 180, 240, and 480 min after isotope infusion, and all urine and feces were collected for 96 h after injection. Seventy percent of labeled circulating steroid was conjugated by 30 min postinfusion. The majority (80.4 +/- 3.2%) of T metabolites were excreted into urine, and 95.0 +/- 0.9% of these were conjugated, 95.8 +/- 0.2% being hydrolyzable with glucuronidase. Seven urinary androgen metabolites, including androstanediol (5 alpha-androstan-3 alpha-17 beta-diol and 5 beta-androstan-3 alpha-17 beta-diol), were identified in glucoronidase-hydrolyzed, ether-extracted Eld's deer urine pools after gas chromatography/mass spectrometry. A double-antibody 125I RIA for 5 alpha-androstanediol-3 alpha, 17 beta-diol,17-glucuronide (3 alpha-diol-G) was validated for unprocessed urine. Longitudinal assessments of urine samples collected from 13 stages for 3 yr revealed biological concordance between fluctuations in urinary 3 alpha-diol-G and serum T, as well as seasonal changes in secondary sexual characteristics. Overall correlation between "same-day" matched serum T and urinary 3 alpha-diol-G was 0.58, (n = 6; p < 0.001). Thus, monitoring urinary 3 alpha-diol-G provides a noninvasive, alternative method for characterizing male endocrine interrelationships in an endangered ungulate species.
