フラグメント分子軌道(FMO)法プログラムABINIT-MPを用いた生体高分子の応用計算の現状をGUIシステムBioSation Viewerの機能とともに紹介する。具体例として、核内受容体とリガンドの相互作用、DNAと転写因子タンパク質との結合などについて、主に転写に関わる分子認識機構を詳細に解析した。フラグメント間相互作用エネルギー、電荷分布、及び軌道相互作用の解析などを通じて、生体高分子の分子認識における各アミノ酸残基の役割や相互作用の様式を明らかにした。またこれらの分子認識には、静電的な相互作用ばかりでなく、分散力に基づくvan der Waals相互作用が重要であることを明らかにし、電子相関を適切に取り入れた量子化学計算が必要となることが具体例を通じて示された。その他にも、光励起特性の解析などの先駆的な研究も進めている。

エストロゲン受容体とエストラジオールとの結合に関する分子メカニズムを解明することを目的とし、エストロゲン受容体にコンピュータ上でアミノ酸変異を導入して、エストラジオールとの結合能をフラグメント分子軌道法を用いて評価した。アミノ酸変異の導入方法の妥当性を検討するために、実験で用いられたのと同じアミノ酸変異を導入して、結合エネルギーを評価し、実験データと比較した。その結果、6種類のアミノ酸変異のうち、3つは定性的に一致し、残り3つは不一致となった。それらの結果の原因を検討した。
