Mediterranean fruit flies (Ceratitis capitata Wiedemann, Diptera: Tephritidae) harbor a community of diazotrophic bacteria in their digestive system. The present study aims to test the hypothesis that bacteria contribute to fly fitness by enhancing copulatory success and egg production in males and females, respectively. After eclosion, flies were fed a full diet containing peptides, sugar and minerals, or a sugar diet, lacking peptides. Subgroups from each diet were fed a mixture of the antibiotics ciprofloxacin and piperacillin. The presence of bacteria, food consumption, weight gain, lipid and protein levels, oviposition in females and copulatory success of males were quantified in the four groups. The antibiotic treatment effectively cleared the gut of bacteria. The relative amounts of food consumed (with or without antibiotics) are similar in all groups. The antibiotics do not inhibit feeding, and their ingestion does not affect dry weight or the amount of protein stored, yet females feeding on the full diet without antibiotics have increased lipid levels. Females fed the full diet produce significantly more eggs than females on the sugar diet, but the presence of bacteria does not affect numbers of eggs produced. However, in the absence of bacteria, the oviposition rate of nutritionally stressed females is significantly accelerated. The presence of bacteria in sugar fed males does not provide them with a mating advantage. Conversely, in males fed a full diet, the presence of bacteria is associated significantly with a shorter latency to mate. It is concluded that, because the bacterial community is present at all stages of the fly’s life cycle, at least some species are effectively transmitted from parents to offspring, and removal of bacteria affects measurable physiological and behavioural parameters related to fitness, the association between bacteria and the medfly is mutualistic.

Effect of bacteria on nutritional status and reproductive success of the Mediterranean fruit fly Ceratitis capitata

One possible control strategy against the olive fly, Bactrocera oleae, the most serious olive crop pest, is the Sterile Insect Technique (SIT) application. However, a number of problems associated with this method remain that decrease the effectiveness of SIT, including the quality of reared insects. Taking the importance of the relationship between the olive fly and bacteria into consideration, the effects of probiotic diets enriched with Pseudomonas putida on B. oleae longevity and fecundity were evaluated. First, we found that the probiotic bacterium, P. putida, is conveyed from diets to the oesophageal bulb as well as to the fly midgut after feeding on the probiotic diet. Subsequently, B. oleae adults fed on either: (a) a standard full protein and sugar diet; (b) a sugar only diet; (c) a probiotic standard full protein and sugar diet; or (d) a probiotic sugar diet. Flies fed on probiotic diets were supplied with an inoculated gel containing P. putida; non-inoculated gel was provided to the flies fed on non-probiotic diets. B. oleae males and females that fed on sugar diets did not survive as long as those that fed on protein diets. A comparison of the longevity of adults fed on full diet and sugar with their respective probiotic diets revealed no significant difference. Males fed on the sugar only diet survived longer than males fed on probiotic sugar diet, and females fed on the full protein and sugar diet survived much longer than females fed on the full probiotic diet. As regarding fecundity, both full diets resulted in a higher number of eggs laid per female. Females fed on the probiotic sugar diet laid a higher number of eggs than females that fed on sugar only. The inoculated gel of the probiotic sugar diet contained a significantly higher quantity of leucine, isoleucine and proline than the non-inoculated gel of the sugar only diet. The possible role of dietary bacteria in relation to functional aspects of olive fly physiology is discussed.

Development of probiotic diets for the olive fly: evaluation of their effects on fly longevity and fecundity

The family Tephritidae (order: Diptera), commonly known as fruit flies, comprises a widely distributed group of agricultural pests. The tephritid pests infest multiple species of fruits and vegetables, resulting in huge crop losses. Here, we summarize the composition and diversity of tephritid gut-associated bacteria communities and host intrinsic and environmental factors that influence the microbiome structures. Diverse members of Enterobacteriaceae, most commonly Klebsiella and Enterobacter bacteria, are prevalent in fruit flies guts. Roles played by gut bacteria in host nutrition, development, physiology and resistance to insecticides and pathogens are also addressed. This review provides an overview of fruit fly microbiome structure and points to diverse roles that it can play in fly physiology and survival. It also considers potential use of this knowledge for the control of economically important fruit flies, including the sterile insect technique and cue-lure baiting.

Tephritidae fruit fly gut microbiome diversity, function and potential for applications.

Characterization of three bacterial symbionts (BC1, BC2 and BC3) of fruit fly Bactrocera tau including morphological, biochemical and 16S rDNA (rrs gene) analysis was done to determine their taxonomic position. Morphological and biochemical characterization placed two bacteria (BC1, Klebsiella oxytoca and BC2, Pantoea agglomerans) into family Enterobacteriaceae and the third one (BC3, Staphylococcus sp.) into family Staphylococcaceae. 16S rDNA gene sequence comparison with the available NCBI database sequences further confirmed the characterizations of bacterial symbionts. Molecular phylogeny of Klebsiella oxytoca and Pantoea agglomerans closely related to the other free living enterobacterial members with 74 to 93% sequence homology (genetic distance 0.000 to 0.085); however, they showed only 74 to 87% similarity with other insect symbionts (genetic distance 0.090 to 0.121). Staphylococcus sp. showed 94% sequence homology with other members of family Staphylococcaceae with the genetic distance of 0.013. Population of these symbionts in adult fruit flies increased exponentially up to the 10th day of adult emergence and thereafter it became almost constant.

Molecular and biochemical characterization of three bacterial symbionts of fruit fly, Bactrocera tau (Tephritidae: Diptera).

Among Australian endemic tephritid fruit flies, the sibling species Bactrocera tryoni and Bactrocera neohumeralis have been serious horticultural pests since the introduction of horticulture in the nineteenth century. More recently, Bactrocera jarvisi has also been declared a pest in northern Australia. After several decades of genetic research there is now a range of classical and molecular genetic tools that can be used to develop improved Sterile Insect Technique (SIT) strains for control of these pests. Four-way crossing strategies have the potential to overcome the problem of inbreeding in mass-reared strains of B. tryoni. The ability to produce hybrids between B. tryoni and the other two species in the laboratory has proved useful for the development of genetically marked strains. The identification of Y-chromosome markers in B. jarvisi means that male and female embryos can be distinguished in any strain that carries a B. jarvisi Y chromosome. This has enabled the study of homologues of the sex-determination genes during development of B jarvisi and B. tryoni, which is necessary for the generation of genetic-sexing strains. Germ-line transformation has been established and a draft genome sequence for B. tryoni released. Transcriptomes from various species, tissues and developmental stages, to aid in identification of manipulation targets for improving SIT, have been assembled and are in the pipeline. Broad analyses of the microbiome have revealed a metagenome that is highly variable within and across species and defined by the environment. More specific analyses detected Wolbachia at low prevalence in the tropics but absent in temperate regions, suggesting a possible role for this endosymbiont in future control strategies.

When 5-day-old laboratory-raised Queensland fruit flies (Dacus tryoni) were fed a dinitrogen-fixing bacterial strain of Klebsiella oxytoca isolated from the crop of a wild fly, acetylene reduction (nitrogenase) activity associated with the flies was detected after 2 to 3 days and persisted for at least 22 days. Flies not fed the dinitrogen-fixing strain were negative for acetylene reduction until 21 days after emergence. Presumably such dinitrogen-fixing bacteria are able to supply some Queensland fruit flies with a small part of their nitrogen requirements, but its importance is unknown.
