The objective of this study was to complete the molecular evaluation of five Acacia species including first by determining the genetic diversity of the plants using the polymerase chain reaction (PCR)-based inter-simple sequence repeat (ISSR) method. This investigation was carried out to assess fingerprint and thus genetic variations among the Acacia species. The ISSR method was used to determine DNA fingerprints for Acacia spp. Eight primers were used, with all primers delivering amplification products. Our data show a total of 71 bands of 70 bp to 2,200 bp were amplified, of which 0.77 demonstrated an average polymorphism information content per primer. Among the eight primers tested, the mean annealing temperature was 48°C and average polymorphism information content was between 0.36 and 0.84. The ISSR primers for the five species of Acacia showed four main groups, with a higher level of similarity between these species. These results indicated ISSR markers provide an efficient alternate for identification via DNA fingerprinting of the genetic relationships in Acacia. PCR-based ISSR represents a powerful method that can provide practical information for the development of molecular markers, molecular cytogenetic techniques, and DNA Fingerprinting for application in an Acacia spp breeding program.

Using DNA Fingerprinting to Detect the Genetic Relationships in Acacia by Inter-Simple Sequence Repeat Markers

The study shows genetic diversity of 38 Vitis vinifera L. cultivars and hybrids originating in North America and Europe, including cultivars selected in Poland, which have not been characterized with the use of DNA markers yet. The agrobiological features of the genotypes selected for testing, indicate that they may be useful for the breeding of new cultivars and grape production. The use of 12 ISSR primers allowed to obtain 94.4% of polymorphism. The polymorphic information content (PIC) value was high and varied between 0.829 and 0.953 with an average of 0.897. The resolving power (Rp) ranged between 3.678 and 8.892 with an average of 6.347. Primers UBC 809, UBC 810, UBC 812, UBC 855, UBC 891 and UBC 810 were found to be highly effective (informative). Similarity coefficient ranged between 0.167 and 1.0, which indicates high degree of diversity of tested  grape cultivars. Tested cultivars were grouped in 3 main clusters; one of them was further divided into 6 subclusters. ‘Pannonia Kincse’ and ‘Danmarpa Polonia’ were not differentiated. Phenotypic differences among those two cultivars suggest that ‘Danmarpa Polonia’ might be a clone of ‘Pannonia Kincse’ and other molecular techniques must be used to differentiate them. Morphological and agrobiological characters of cultivars support the results obtained by ISSR markers.

Assessment of genetic diversity and relationships among grapevine cultivars originating in Central and Eastern Europe and North America using ISSR markers

In the present work, 43 seedless grape (Vitis vinifera) varieties were evaluated using 14 informative ISSR primers. A total of 119 bands were scored, out of which 79% were polymorphic. The level of polymorphism varied according to the primers which was evident also from different Resolving Power values of the individual primers. Based on the DNA fingerprint data of only three primers (UBC 857, 888 and 890), all the varieties except one could be identified. UPGMA cluster analysis revealed high degree of similarity among the varieties. Distinct subgroups based on the berry colour were observed. No intravarietal differences were detected. Overall grouping was in accordance with morphological characters and pedigree, proving usefulness of ISSR markers to assess genetic relationships among grape varieties.
